BACKGROUNDThe natural enviroment of plateau(more than 3000 meters)is more hostile,It characterizes with hypoxia, severely cold,high radiation and big temperature difference by day, etc.When the general population from low altitude area enter this area, due to the enviroment induce lower air pressure, forming oxygen partial pressure difference, the body appear low oxygen reaction.First,the respiratory system make adaptability adjustment, the frequency of breathing is added fast, then heartbeat rate and cardiac output increases accordingly, so increasing blood flow improve ability of transports oxygen, improve hypoxia of the body. In fact,even if the body make it to adjust accordingly, the physiology in plateau enviroment is lower than low altitude, the affected systems include resiratory, circulatory, nervous, skeleton and digestion et al. At present, the studies about high altitude medicine focus on resiratory, circulatory and nervous system in the domestic and aboard, compared with them, the digestion system about high altitude medicine have little research. With the opening of the Qinghai-Tibet railway, the population from low altitude to the plateau will surely increase, so in theory, the number of enteric diseases also rise accordingly. Such as diarrhoea, anorexia symptoms etal was simply believed "unacclimatized", there is actually close relationship between "unacclimatized" and intestinal barrier dysfunction. Not only nutrient absorption and digestion, but the gastrointestinal tract serves as a potent barrier, fluid secretion and immunization et al. The importance of intestinal barrier for the body is being knowed more and more scholars. The protein as the most importance structure protein of tight junction is the hot spot of research. The number of living microbes in humanbeing's gut are beyond the cells of itself, If the intestinal mucous barrier is damaged and gaps of tight junctions are magnified, which can help harmful material such as bacteria to translocate. It must cause intestinal endotoxemia(IETM) and aggravated the jury of intestinal mucous barrier, forming vicious cycle. The worst consequences are multiple organ dysfunction syndrome (MODS),systemic inflammatory response syndrome (SIRS), threatening life. So this study was to estibilish model of hypoxia's rat on high altitude and the rats were afford-ed galacto oligo saccharides(GOS), in order to study protein occludin of intestinal mucous.AIMSTo investigate relationship between the effect of occludin expression of intestinal mucous and damage of intestinal mucous barrier in the plateau,we estibilish model of hypoxia's rat on high altitude; in order to demonstrate the value of GOS, we afford the GOS to rats. Immunohistochemical methods were used to measure the expression levels of occludin; Real time RT-PCR methods were employed to quantite the occludin mRNA; The correlations between expression levels of occludin and occludin mRNA were analyzed.MATERIALS AND METHODSthe wistar rats of 48 were purchased from Breeding Laboratories of lanzhou university medical college. Forty-eight models rats in radical plateau hypoxia environment were established(3 848m). These rats were randomly assigned to hypoxia control group(n=24) and probiotics group(n=24), then were randomly divided into six groups(n=8) at 2d,4d,6d point-in-time. Animal were narcotized by chloraldurate abdominal injection and their ileums were obtained by surgery. A part of ileums,Immunohistochemical methods were used to measure the expression levels of occludin; and other's,Real time RT-PCR methods were employed to quantite the occludin mRNA; The correlations between expression levels of occludin and occludin mRNA were analyzed in prebiotics group, the effect of gos expression may be protective mechanism in mucuosa.RESULT (1) We have successfully established the model of hypoxia's rat on high altitude(3 848m)by Mountain ATV.(2)To demonstrate expression levels of occludin 2d>4d>6d in the hypoxia control group and expression levels of occludin 2d<4d<6d in the prebiotics group, the comparison of theirs indicates that expression levels of occludin higher in the prebiotics group than in the hypoxia control group contemporaneity(P<0.05)(3) The result of occludin mRNA quantiton by Real time RT-PCR method coincide with the result of expression of occludin by Immunohistochemical methods.they have a significant positive correlation (R=0.9553,P<0.01), It suggests that the prebiotics can increase expression levels of occludin by incresing quantiton of occludin mRNA and effect of improving intestinal mucous barrier.CONCLUSIONSThe study demonstrate the hypoxia causing demage for intestinal mucous by establishing the model of hypoxia's rat on high altitude(3 848m). Probiotics have a value of treatment that improve intestinal mucous impairment being caused hypoxia in plateau. It provides the new ideas and methods of treatment for the future research of the plateau gastroenterology.
