Phellinus baumii is one sourse of "Sang huang", which is a Traditional Chinese Medicine used for a long time.Recent research has shown that components extracted from P. baumii have antitumor activity, antioxidant activity anti-hepatic fibrosis function, and immunomodulatory properties.This paper mainly studies on the chemical constituents, antibacterial activity in vitro, anti-tumor activity in vivo and extraction technology of flavonoids from the fruiting of this fungus.In order to promote further utilization of this medicinal fungus, and providing some basic datas for the comprehensive development of "Sang huang".The main results were as follows:1. The dried fruiting body of Phellinus baumii were crushed,then extracted by by petroleum ether (30-60), chloroform, ethyl acetate, acetone, and methanol.Five extracts were obtained, and nine compounds were isolated from the extracts by normal phase silica gel and Sephadex LH-20 chromatography. Their structures were elucidated by spectroscopic methods include 1H-NMR,13C-NMR,DEPT and MS. They were identified as ergosta-7,22-diene-3-one, ergosta-4,6,8,22-tetraen-3-one, ergosterol, ergosterol peroxide,2-(4-hydroxyphenyl)ethyl-1-dodecyloctade-canoate, hispolon,4-(3,4-dihydroxyphenyl)-3-buten-2-one, hispidin and cerebrosideB.ergosta-7,22-diene-3-one,ergosta-4,6,8,22-tetraen-3-one,4-(3,4-dihydroxyphenyl)-3-buten-2-one were first found in this fungus, and 2-(4-hydroxyphenyl)ethyl-1-dodecyloctade-canoate was first found in Fungi.2. The antibacterial activity of extracts of Phellinus baumii in vitro were tested by K-B paper disk method and broth dilution method. Petroleum ether extract showed antibacterial activity against Staphyloccocus aureus ATCC6538 and Escherichia coli ATCC8099 in a concentration-dependent manner, the extracts showed the highest inhibition ratio at the concentration of 40.96mg/mL, inhibition ratio were 35.33% and 40.49%. Different concentrations showed inhibitory effect against Bacillus subtilis, the highest inhibition ratio was 10.96%, at the concentration of 40.96mg/mL.The minimum inhibitory concentration of petroleum ether extract against the bacterias were 2.048 to 4.096mg/mLChloroform extract showed antibacterial activity against E. coli ATCC8099, S. aureus ATCC6538 and B.subtilis. The highest inhibition ratio were 15.09%,44.22% and 10.13% at the concentration of 40.96mg/mL. The minimum inhibitory concentration of chloroform extract against E.coli ATCC8099 was 2.048mg/mL.The minimum inhibitory concentration against S.aureus ATCC6538 and B.subtilis were higher than 4.096mg/mL.EtOAc extract showed antibacterial activity against S.aureus ATCC6538, E.coli ATCC8099 and B.subtilis in a concentration-dependent manner.The highest inhibition ratio were 32.04%,37.7% and 8.90% at the concentration of 40.96mg/mL.The minimum inhibitory concentration of EtOAc extract against E.coli ATCC8099, S.aureus ATCC6538 and B.subtilis were 1.024mg/mL,4.096mg/mL and 4.096mg/mL.Acetone extract showed antibacterial activity against E.coli ATCC8099, S.aureus ATCC6538 and B.subtilis in a concentration-dependent manner, the highest inhibition ratio were 32.98%,33.50% and 11.34% at the concentration of 40.96mg/mL.The minimum inhibitory concentration of acetone extract against S.aureus ATCC6538, E.coli ATCC8099 and Bacillus subtilis were 1.024mg/mL,0.512mg/mL and 2.048mg/mL.Methanol extract inhibited E.coli ATCC8099 and B.subtilis in a concentration-dependent manner, the highest inhibition ratio were 34.69% and 6.81% at the concentration of 40.96mg/mL. High concentration showed inhibitory effect against S.aureus ATCC6538 inhibited, the inhibition ratio were 9.48% and 2.37%. The minimum inhibitory concentration of methanol extracts against S.aureus ATCC6538 and E.coli ATCC8099 was 4.096mg/mL and 2.048mg/mL, the minimum inhibitory concentration against B.subtilis was higher than 4.096mg/mL.3. Studied on anti-tumor activity in vivo, we found petroleum ether extract inhibited solid tumor growth on mice bearing implanted H22 tumor cells. The inhibition ratio reached 19.76% (100mg/kg/d). Spleen index were significantly increased(P<0.05), compared with the control group and CTX group. Methanol extract (1000mg/kg/d)inhibited the solid tumor growth too, the inhibition ratio reached 22.12%, compared with the control group, the spleen index was significantly increased(P<0.05). The results showed that petroleum ether and methanol extracts of fruiting body of Phellinus baumii inhibited the growth of the solid tumor, and showed a good role in promoting immunity.4. Extraction technology of flavonoids fom Phellinus bauntii was studied by single factor test and orthogonal test. Effects of ethanol-water solvent, extraction temperature, extraction time and ratio of materials to liquid were investigated. The optimum extraction condition according to the L9(34)orthogonal test were obtained:temperature 80℃, extraction time 8 hours, ratio of materials to liquid 1:20 and ethanol concentration 70%.By the above conditions, the extraction of flavonoids up to 15.5027±0.3318mg/g.
