Studied On The Cardiovascular Effects And Chemical Components Of Ilex Rotunda Thunb Saponins

To seek an effective medicine for the treatment of cardiovascular disease, we used the traditional Chinese medicine-Ilex Rotunda Thunb. as raw materials and carried out a systematic study on the chemical constituents of Ilex Rotunda Thunb. saponins by applying to macroporous resin, silica gel columns, ods columns, prep-HPLC and recrystallization. Three compounds were isolated from the saponins of Ilex Rotunda Thunb. and on the basis of chemical methods and spectroscopic analysis including UV, IR,1H-NMR,13C-NMR, MS, their structures were elucidated. At the same time, we formulated the quality standard of these compounds by applying to HPLC. Those offered a theoretical basis for the further development of Ilex Rotunda Thunb. as a cardiovascular drug.In addition, we investigated the stability of Ilex Rotunda Thunb. saponins sample, including character, identification, inspection, determination, hygiene and other projects, the results showed that:in the study period, the stability of the samples were found to be no obvious changes, and the accelerated test of Ilex Rotunda Thunb. saponins also be compliance.Ilex Rotunda thumb, is a herb of Aquifoliaceae, alias Ovateleaf Holly Bark, which is our southern common herb with bark or leaves used medicinally. The pharmacological effects of it are heating or colding blood, reducing inflammation, and relieving pain. And it is used as a common herb with the treatment of infectious diseases and stomach disease in early period. In recent, there are literature gives reports that the pharmacological effect of Ilex rotunda thumb, on cardiovascular is more obvious, and the water extract of Ilex rotunda thumb.leaves can add to the coronary plasma flow of cavy heart in vitro, which is effective like Salvia miltiorrhiza injection. The ethanol extract of Ilex rotunda thumb.can be step-down quickly and short-acting on rats. The butanol extract of Ilex rotunda thumb.containing the active ingredient of antiarrhythmic and protection of myocardium with a shortage of oxygen and blood, and this active ingredient also can be step-down on rats, especially diastolic blood pressure. That has a positive significance for the treatment of hypertension and coronary heart disease.At present, Ilex Rotunda Thunb. usually used for the treatment of gastrointestinal diseases, and the development and utilization of it is also very limited beacause of less demanded. In our study, three compounds which against cardiovascular disease were isolated from Ilex Rotunda Thunb., their structures were elucidated as holly glycosides with habitat, syringin and phloroglucinol. That can make full use of the herb and provide a new application market for the massive planting, and it will promote its cultivation, production, processing and related industries.
