objective:To establish a layered coculture modeI of BMSCs and NPCs invitro,and to observe the NPCs effect of on the differentiation of BMSCs.To demenstrate the regenerative effects of transplanting with tissue engineering nucleus pulposus to the degenerated intervertebral disc, and to provide a new approach for the treatment of the degenerative intervertebral disc disease.method:the culture,identincation and choose of the rabbit BMSCs and NP cells,BMSCs'and NP cells'biological features were research;To establish a layered coculture model of BMSCs and NPCs invitro,The BMSCs were researched by inverted phase comrast microscope,light microscope,Immunohistochemistry and RT-PCR,and induce it differentiate into osteoblast. isolated and cultured rabbit bone mesenchymal stem cells in vitro,cultured the third generation BMSCs in Seaweed salt hydrogels.To study it by inverted phase comrast microscope,Immunohistochemistry and use CM-Dil. Deteced the histocompatibility of seaweed salt gel for the seed cells. To establish the disc degeneration model by needle puncture injury disc. Transplantated the tissue engineerning nucleus pulposus into the degenerative disc.after raised for 7,14,28,56 day,take examine the rabbit model by MRI,and sacrificed ,isolated disc and observed by histological stain and gene express essay,and respectively compared the results with untreated degenerative dics and normal disc to identify whether the complex could repair the degenerative disc.consequence:The isolation, culture and identification of BMSCs and NPs, and the research results demonstrate the superiority of BMSCs as seed cells, in the three-dimensional co-culture system,cultured with NP cells,BMSCs differentiate into nucleus pulposus cells,and express the biological characteristics of nucleus pulposus cells. To establish tissue engineering nucleus pulposus by the seed cells,and transplantate it into the rabbit disc degeneration model,the result show it that the tissue engineering nucleus pulposus can repair the degeneration of intervertebral disc.conclusion:1.The MSCs which its rescoure is widespread,is easy to be isolated and directed differentiate under certain conditions are the most suitable resource for the tissue engineering seed cells.2. Seaweed salt gel has favourable histosompatiblity.3.The tissue engineering nucleus pulposus can repair the degeneration of intervertebral disc.
