Purpose:To observe the influence of the expression of intercellular gap junctions protein CX43 with Thalidomide in the prostate cancer cells line ,and investigate the possible anti-tumor mechanism of thalidomide.And provide further theoretical basis for the treatment of prostate cancer with Thalidomide.Method and Materials:The different concentration of thalidomide (0ug/ml,12.5ug/ml,25ug/ml,50ug/ml,100ug/ml)were intervened prostate cancer PC-3 cells in logarithmic growth for 24 hours and 48 hours. Using the method of the real-time fluorescence quantitative PCR to detect the expressions of Cx43 mRNA in cells and the Western Blot to test the expression of Cx43 protein in cells.Scrape-loading dye transfer method was applied to detect gap junction intercellular communication.Results:Real time-PCR and Western Blot detection results: results:There was different improvement of the CX43 mRNA and its protein expression in different concentration groups of PC-3 cells after being treated thalidomide.Thalidomide obviously improved the CX43mRNA and its protein expression in PC-3 at 100ug/ml after treating 48h, and there was significantly statistical significance between before and after treatment(P<0.05).Scrape-loading dye transfer results: the function of GJIC for PC-3 prostate cancer cells have different levels of restoration or enhancement, with THD enrichment, LY dye transmission range increases by a dose dependent manner and time-dependent effects of dependency.Conclusion: The THD can increase the levels of mRNA and protein which Cx43 gene expressd and promote the function of GJIC in prostate cancer PC-3 cells, which may be one of the mechanisms of its anti-tumor.
