The patterns of efficient components recognition and quality control for Traditional Chinese medicine (TCM) have been the difficult and hot topic for TCM modernization. This study a new method for the screening and analysis of bioactive components and quantity control of TCM was developed and named "Knock-out & Knock-in". In this paper, this pattern was used for screening and analysis of the key bioactive components in Flos Lonicerae Japonicae which is a famous traditional Chinese medicine (TCM). Then the negative samples of the key bioactive components were used as background for determining the effect of TCM. In detail, the key bioactive components were added into the background at different doses to study their dose-effect relationships in background. On the basis of quantitative study on key components, the reasonable and precise quantitative standard could be established. Results as following:1.Different chemical parts of Flos Lonicerae Japonicae were prepared by column chromatography. The "Knock out & Knock in " samples were prepared by the proportion of paste-forming rate of the chemical parts prepared. The "Knock out & Knock in " samples were credible.2.Based on Constituent Knock-out/Knock-in, the key bioactive components of Flos Lonicerae Japonicae were screened. The anti-bacterial effect of different samples were compared by thermodynamics. The anti-viral effect of different samples were compared by studying the inhibition activity of the samples to enterovirus (EV71) in vitro.isochlorogenic acids were identified to be the key bioactive components.3.Based on Constituent Knock-out/Knock-in, the dose-effect relationship of isochlorogenic acids in the background of the negative samples of it. The quantity-antibacterial relationship of "Knock-in" isochlorogenic acids were researched by thermodynamics. The quantity-antiviral relationship of "Knock-in" isochlorogenic acids were researched by anti-influenza virus (H1N1) in vivo. On the basis of dose-effect relationship study on "Knock-in" isochlorogenic acids, the reasonable and precise quantitative standard of Flos Lonicerae Japonicae was established.In the paper Flos Lonicerae Japonicae was selected as mould which is effective and universal in clinic usage. This research has representation, and the conclusion can be popularized and applied. And patterns of efficient components recognition and quality control for TCM based on constituent Knock-out/Knock-in are forward-looking. The patterns can be applied for the TCM which active substance is not clear or/and quality control establishment is difficult.
