Object: Many studies indicate that tumor invasion and metastasis are oncogenes and tumor suppressor genes involved in the regulation of a very comPlex Process, in this Process, involving many factors, such as extracellular matrix and basement membrane degradation and the abnormal number of adhesion molecules Expression, integrin family, the growth factor family and so on.Some studies have reported associated protein meningioma (Meningioma associated protein, MAC30) is the insulin-like growth factor binding protein IGFBP member of the family, in oral squamous cell carcinoma, colorectal cancer, breast cancer, lung cancer, skin cancer and other cancer tissues overexpressed , also found that lymph node metastasis group than in the group without lymph node metastasis positive rate of MAC 30 was significantly increased, suggesting MAC30 protein expression in tumor tissue increased with tumor progression and metastasis prognosis. However, esophageal squamous cell carcinoma MAC30 on protein expression and clinical significance has not been reported at home and abroad. COX-2 gene is located on human chromosome 1q25.2-25.3, length of 8.3Kb, contains 10 exons. COX-1 is a COX-2 isomers, in the most positive expression of tissue structure; and COX-2 expression in normal circumstances is strictly limited, and only in the cells are stimulated, it can be rapidly induced, rapid de novo synthesis, involved in many pathological processes, while producing prostaglandins can cause inflammation and pain. Recently, more and more evidence that, COX-2 expression is also associated with tumor occurrence and development related. Of MAC30 protein and COX-2 protein in esophageal squamous cell carcinoma significance and relevance, little has been reported at home and abroad. The PurPose of the experiment through the normal esophageal mucosa and esophageal squamous cell carcinoma MAC30 Protein, COX-2 Protein expression in the detection, of MAC30 protein, COX-2 protein expression in esophageal squamous cell carcinoma, the role of the development process as well as in esophageal squamous cell carcinoma invasion and metastasis of the correlation; were observed MAC30 Protein, COX-2 Protein in esophageal squamous cell carcinoma and age, sex, tumor gross morphology, histological grade and clinical stage, lymph node metastasis; and compare MAC30 Protein, COX-2 Protein expression in esophageal squamous cell carcinoma and relevance of the relationship between the two, in order to determine the clinical tumor invasion, metastasis and Prognosis of two new biological markers.Methods:Normal esophageal mucosa tissue samples collected 22 cases of esophageal squamous cell carcinoma in 68 patients using immunohistochemistry (Immunohistochemistry, IHC) SP method detected in normal esophageal mucosa and esophageal squamous cell carcinoma MAC30 protein and COX-2 protein expression. Statistical software was used SPSS18.0 processed toα=0.05 level for the test, when P<0.05 when statistically significant. The Positive rate calculated count data, the sample was used to compare between the rate of chi-square test, correlation test between the two variables using Spearman rank correlation analysis.Results:1 The Positive expression rate of protein MAC30 in esophageal squamous cell carcinoma is 51.6%(32/62), is apparently higher than that in normal esophageal mucosa tissue with 0%(0/26). The difference between the two is significant (P<0.05). There is no correlation between the expressions of MAC30 and the patient's gender and age, gross morphology of tumor and its differentiation. The positive expression rate of protein MAC30 is 71.4% (15/21) in squamous cell carcinoma tissue with lymph node metastasis, but the positive expression rate of protein MAC30 is 41.5% (17/41) in the same cases without lymph node metastasis. The difference between the two is significant (P<0.05). The positive expression rate of Protein MAC30 is 55.4% (31/56) in moderate and advanced squamous cell carcinoma tissue, but the positive expression rate of protein MAC30 is 0% (0/6) in early-stage squamous cell carcinoma tissue. The difference between the two is significant (P<0.05).2 The positive expression rate of COX-2 in esophageal squamous cell carcinoma is 38.7% (29/62), is apparently higher than that in normal esophageal mucosa tissue with 0% (0/26). The difference between the two is significant (P<0.05). There is no correlation between the expression of COX-2 and the patient's gender and age, clinical stage, gross morphology of tumor and its differentiation. The positive expression rate of COX-2 in squamous cell carcinoma tissue with lymph node metastasis, i.e.66.7% (14/21), is apparently higher than that in the same cases without lymph node metastasis 46.8% (15/41). The difference between the two is significant (P<0.05).3 There is correlation between the positive expressions of MAC30 protein and COX-2 protein in 68 detected cases of esophageal squamous cell carcinoma. They are positively correlated (P<0.05), and correlation coefficient is 0.324.Conclusions:1 The result that the expression of MAC30 in esophageal squamous cell carcinoma is apparently higher than that in normal esophageal mucosa tissue suggests that protein MAC30 is relevant to the tumorigenesis of esophageal squamous cell carcinoma. The result that the expression of MAC30 is obviously relevant to lymph node metastasis suggests that protein MAC30 is possibly relevant to the invasion, development and metastasis of esophageal squamous cell carcinoma. Thus, MAC30 could be as a tumor marker for the Prediction of invasion and metastasis of esophageal squamous cell carcinoma and it has a certain prompting significance for Prognosis.2 The result that the expression of COX-2 in esophageal squamous cell carcinoma is apparently higher than that in normal esophageal mucosa tissue suggests that protein COX-2 is possibly relevant to the tumorigenesis of esophageal squamous cell carcinoma. What's more, the result that the expression of COX-2 with lymph node metastasis is apparently higher than that without lymph node metastasis suggests that protein COX-2 is closely relevant to the invasion and metastasis of esophageal squamous cell carcinoma. And the detection of the expression of COX-2 possibly has an important significance on estimating the patient's prognosis.3 There is no correlation between the expressions of MAC30 and COX-2 and the patient's gender and age, tumor differentiation and tumor gross morphology of esophageal squamous cell carcinoma.4 There is correlation between the abnormal expressions of protein MAC30 and COX-2 in esophageal squamous cell carcinoma, and they are positively correlated.
