Background Psoriasis is a common, chronic and recurrent, inflammatory skin disease. Genetic and environmental factors play important roles in the pathogenesis of psoriasis. Psoriasis vulgaris (PV) is the most common clinical type. We found a distinguished correlation of SNP rs999556 and SNP rs3762999 in ANXA6 gene with psoriasis vulgaris in Chinese Han Population by psoriasis genome-wide association study (GWAS) and a multistage replication study.Objective Based on stratifying analysis of the age of onset, family history and clinical types, we compared the correlation of SNP rs999556 and SNP rs3762999 in the ANXA6 gene with some clinical features of psoriasis vulgaris in the Chinese Han population in order to further explore the genetics pathogenesis.Methods We analyzed the distributions of genotype and allele of SNP rs999556 and SNP rs3762999 in ANXA6 gene in 7425 cases and 11208 normal controls, which come from the data of the psoriasis genome-wide association study (GWAS) and a multistage replication study. The social science statistical software package SPSS 10.0 was used for statistical analysis of data.Results 1. The genotype frequency of SNP rs999556 and SNP rs3762999 in ANXA6 gene was of statistical significance between the cases and the controls(P=5.22×10-21;P=1.41×10-16), there was a similar trend for the allelic frequency of SNP rs999556 and SNP rs3762999 in ANXA6 gene(P=2.63×10-22;P=1.27×10-17). 2. The genotype and allelic frequencies of SNP rs999556 were statistical significance between the pediatric age of onset cases and controls(P=4.56×10-6;P=8.88×10-7), there was a similar trend of SNP rs3762999 (P=3.02×10-4;P=5.97×10-5). The genotype and allelic frequencies of SNP rs999556 were also statistical significance between the adult age of onset cases and controls (P=9.08×10-20;P=8.68×10-21), there was a similar trend of SNP rs3762999(P=3.67×10-16;P=4.59×10-17). However, there weren't statistical significance between the pediatric age of onset and adult age of onset cases of SNP rs999556(P=0.22;P=0.26), there weren't statistical significance of SNP rs3762999(P=0.26;P=0.17). 3. The genotype and allelic frequencies of SNP rs999556 in ANXA6 gene were statistical significance between the familial cases and controls(P=8.34×10-10;P=1.18×10-10), there was a similar trend of SNP rs3762999(P=3.56×10-8;P=8.57×10-9). The genotype and allelic frequencies of SNP rs999556 were also statistical significance between the sporadic cases and controls(P=9.74×10-17; P=6.34×10-18), there was a similar trend of SNP rs3762999(P=4.79×10-13;P=4.84×10-14). However, there weren't statistical significance between the familial and sporadic cases of SNP rs999556(P=0.74;P=0.49), there also weren't statistical significance of SNP rs3762999(P=0.65;P=0.47). 4. The genotype and allelic frequencies of SNP rs999556 in ANXA6 gene were statistical significance between the acute guttate cases and controls(P=4.14×10-7;P=1.40×10-6), there was a similar trend of SNP rs3762999(P=4.89×10-5;P=5.86×10-5). The genotype and allelic frequencies of SNP rs999556 were also statistical significance between the chronic plaque cases and controls(P=3.66×10-20;P=2.80×10-21), there was a similar trend of SNP rs3762999(P=3.61×10-16;P=3.12×10-17). There was statistical significance between the acute guttate and chronic plaque cases of the genotype frequency of SNP rs999556(P=0.01), there weren't statistical significance of the allelic frequency of SNP rs999556(P=0.15); and there weren't statistical significance between the acute guttate and chronic plaque cases of the genotype frequency and allelic frequency of SNP rs3762999(P=0.07; P=0.12).Conclusion 1. The SNP rs999556 and SNP rs3762999 in ANXA6 gene is associated with the susceptibility of psoriasis vulgaris in the Chinese Han population. 2. The SNP rs999556 in ANXA6 gene may be associated with clinical type of psoriasis vulgaris. 3. The SNP rs999556 and SNP rs3762999 in ANXA6 gene may be not associated with age of onset of psoriasis vulgaris and family history of psoriasis vulgaris, the SNP rs3762999 in ANXA6 gene may be not associated with clinical type of psoriasis vulgaris. |
