Objective To prepare Hydroxybutyl chitosan (HBC) and study its characteristics such as water-solubility, thermosensitive, biocompatibility and so on. To investigate the feasibility of using HBC as tissue engineering materials.Methods The HBC were prepared by chitosan chemically modified. We change the reaction time and temperature to find the optimal preparation conditions.Furthermore, increasing the temperature of water bath heating to get the HBC hydrogel.The structure of the HBC was characterized by IR and scanning electron microscopy(SEM). The DS of HBC was calculated by elemental analysis.The effects of reaction conditions on the incipient gelation temperature (IGT) were studied. MEF were cultivated and the biocompatibility of HBC is investigated by using the method of the MTT assay.Results All the four HBC samples can dissolve in water, HBC4 which was reacted 14 hours at 55℃have better water-solubility. DS of the four groups of HBC were (1.19±0.04),(1.33±0.03),(1.42±0.02) and (1.73±0.01).HBC solution can rapidly transform to semisolid at body temperature and revert to its solvated state at low temperature.IGT of HBC gels presented in this study are ranging from 20 to 34℃. MEF cultured with 48h leachates of HBC are investigated by the MTT assay and RGR is calculated and the cytotoxicity is graded by generally accepted standard. Conclusion:A novel thermosensitive hydrogel HBC was prepared successfully which could be transformed into gelation at 37℃. Vitro cytotoxicity test has shown HBC hydrogel has good biocompatibility which provides possibilities and foundations for the further research.
