Objective To study serum level of retinol binding protein 4(RBP4) and related factors in patients with abnormal glucose metabolism.Methods (1) Incipient abnormal blood glucose and normal people among Chinese Han population in Chongqing area were collected who meet the inclusion conditions. Totally 80 subjects, 36 male and 44 female. Anthropometric parameters, blood pressure, lipid profile, insulin levels etc. were measured, as well as OGTT.Fasting serum RBP4 concentrations were determined by ELISA method. T2DM and IGR were diagnosed according to WHO diagnostic criteria. (2) All the subjects were divided into three groups:T2DM group, IGR group and normal glucose tolerance(NGT) group. The difference between clinical and biochemical indicators were compared among the three groups of their anthropometric parameters,blood glucose , the levels of insulin, serum RBP4 concentrations etc., and the correlation of indexes and the serum RBP4 was analyzed by SPSS statistical software.Results The serum level of RBP4 of T2DM and IGR groups was higher than that of NGT group, but there was no statistically difference in the RBP4 level between these two groups. The risk factors to affect the occurrence of T2DM were BMI,WHR,hypertention,dyslipidemia,HOMA2IR,HOMA2%B and the level of RBP4.But RBP4 was not a major risk factor to affect the occurrence of T2DM.In abnormal blood glucose (T2DM/IGR) group, multiple correlation and regression analysis showed that FINS, HOMA2IR, and SBP were positively correlated with RBP4, but there was no indicator showing the main factors affecting serum RBP4 level. The level of serum RBP4 increased in abnormal blood glucose group combined with hypertension, but there was no difference when combined with obesity and dyslipidemia.Conclusions The serum level of RBP4 of the people of incipient T2DM and IGR was higher than that of normal people,as well as some relation with T2DM and its related clinical parameters suggest that RBP4 possibly plays a role in the occurrence and development of T2DM . Totally 335 subjects, 184 T2DM subjects and 151 normal subjects.They had no genetic relationship. T2DM were diagnosed according to WHO diagnostic criteria.(2)A technology of matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) was applied to detect the polymorphism of rs17484721 and rs3758539 locus in RBP4 gene .At the same time ,the datas were analyzed by SPSS statistical software.Resultsâ‘ There was no statistically significant difference between diabetes and normal control group for genotypes and allele frequency of polymorphism of rs17484721 and rs3758539 locus in RBP4 gene (p>0.05).â‘¡In the diabetic group, the value of the HOMA2IR of AA genotype carrier of rs17484721 was significantly higher than that of other genotypes (ANCOVA=5.181,P=0.024),the value of the fasting blood-glucose of the GA+AA genotype carrier of rs3758539 was significantly higher than that of GG genotypes(ANCOVA=6.017,P=0.015).â‘¢There was statistically difference between dyslipidemia group and normal group for genotypes of rs17484721 in RBP4 gene (p=0.012).AA genotype carrier of rs17484721 was easier to have dyslipidemia than GA+AA genotype carrier.Conclusions RBP4 Gene polymorphism of Chinese Han population may be associated with type 2 Diabetes.
