Objective:To investigated the effect of near-infrared fluorescent quantum dots (QD800)on the proliferation, adherence, invasion and chemotaxis of human tongue squamous cell Tca8113 in vitro. To observe the effect of the QD800 on growth, proliferation, apoptosis and lymphatic metastasis of human tongue squamous cell Tca8113 and Kunming mouse lymphatic high metastatic squamous cell U14 in vivo.Methods:①Different concentrations of QD800 was applied to Tca8113 cells, and the cell growth of control and three test groups were compared respectively;②Tca8113 Cell suspension was co-cultured with QD800, The clone formation efficiency of Tca8113/QD800 and Tca8113 cells were compared respectively;③Tca8113 cells were labeled by QD800 (Tca8113/QD800) , then transwell chambers and wash way were used to dectect the difference of invasion and metastasis ability between Tca8113/QD800 and Tca8113 cells.④Tca8113 and U14 cells were labeled by QD800(Tca8113/QD800, U14/QD800).Tca8113/QD800 and U14/QD800 were inoculated respectively subcutaneously into nude mice and Kunming mice.The tumor formation of Tca8113/QD800 and Tca8113,U14/QD800 and U14 was observed and compared in vivo.The proliferation and apoptosis of Tca8113/QD800and Tca8113,U14/QD800 and UI4 cells from tumors formed in vivo were analyzed bv flow cvtometry.⑤U14/QD800 and U14 were inoculated into the buccat Ixlueosa of Kunming mice to establish the cervical lymph node metastasis model of bucca1 cancer.The cervica1 lymph node metastatic ability of U14/QD800 and U14 was comparedResults:①Different concentrations of QD800 showed no negative effects on growth of Tca8113 cells;②The clone formation efficiency and the ability of invasion, attachment, motive were not significantly different between Tca8113/QD800 and Tca8113 cells (P>0.05) .③The tumor weight and volume of Tca8113/QD800 and Tca8113,U14/QD800 and UI4 in vivo were not significantly different (P>0.05),the cell proliferation index and apoptosis index of Tca8113/QD800 and Tca8113,U14/QD800 and UI4 in vivo were not significantly different (P>0.05) .④The cervical lymph node metastasis rate of U14/QD800 and UI4 buccal cancer were not significantiy different(P>0.05) .Conclusion:①The growth, proliferation, invasion, and metastasis ability of Tca8113 cells were not affected by NIRF-QD800.②The QD800 do not affect the Tca8113 cells and UI4 cells'capacities to grow, proliferate, and metastasize. Our study provides scientific bases for the application of NIRF-QDs in non-invasive visualization in vivo imaging for cancer cells growth and metastasis.
