ObjectiveTo investigate the effect of extracellular signal-regulated proteinkinase (ERK) 1/2 on the expression of bilirubin induced nucleartranscription factors (NF-кB) and the possible mechanism of ERK1/2signal pathway preventing bilirubin neurotoxity in hippocampal neurons invitro.MethodsHippocampal neurons cultured in vitro were randomly divided intocontrol group, biliruibin group, PD98059 (MEK/ERK inhibitor)+bilirubingroup and erucin (ERN,ERK activator)+bilirubingroup. Neuronal viabilitywas tested by modified MTT. The apoptosis rate was tested by dualfluorescent staining with Hoechst33342/PI and verified by dual fluorescentstaining with Annexin V﹣FITC/PI. The expression of NF-кB inhippocampal neurons were detected by immuocytochemistry.Results(1)Differences of the neuronal relative viability among groupswere significant (P < 0.01): control group 100% > ERN+bilirubin group (80.020±8.552)% > biliruibin group(67.546±3.241)% > PD98059+bilirubin group(50.591±3.981)%.(2)Differences of the apoptosis rates of hippocampal neurons bydual fluorescent staining with Hoechst33342/PI were significant(P < 0.01): control group (4.7957±1.109) % < ERN+bilirubingroup (10.429±1.148) % < bilirubin group (21.2773±2.039) %< PD98059+bilirubin group (31.6532±2.596) %.(3)Differences of the apoptosis rates of hippocampal neurons bydual fluorescent staining with Annexin V-FITC/PI weresignificant (P < 0.05): control group (6.859±1.559) %
