PartⅠThe efficacy and mechanism to allergic asthmatic mouse with different dose of dermatophagoides farinae vaccine Objective To investigate the efficacy and mechanism to allergic asthma mouse with different dose of dermatophagoides farinae (der f) vaccine.Methods 32 female C57BL/6 mice were randomly divided into 4 groups after setting up the allergic asthma mode with the extract of der f: normal sodium control group(group A), low dose der f group(group B),middle dose der f group(group C), high dose der f group(group D). The group A was treated with normal sodium all along, other groups were treated intraperitoneally with corresponding dose der f. After treatment, airway reactivity of mice was assayed. The total cell numbers and composition of BALF were determined. The phenotype and the expressing of costimulatory molecules of dendritic cells were detected with flow cytometry. The levels of IL-4, IFN-γin BALF and the level of IL-12 produced by dendritic cells were also measured by ELASA.Results The airway hyperreactivity , the maturity of DC, the total cell number and composition of BALF were significant reduced in group C and D, and there is a great increase of IFN-γand IL-12 in group C(P<0.05).Conclusion The der f vaccine has a great efficacy to allergic asthma, but the therapeutic effect has a close relationship with the therapeutic dose. Dc plays a great role in the immunotherapy .PartⅡInfluence of specific immunotherapy on dematophagodides pteronyssinus specific IgG4 and pulmonary function in allergic asthmaObjective To investigate the therapeutic effect , including the pulmonary function, airway reactivity, dermatophagoides pteronyssinus(Der p)specific IgG4,and the Der p specific IgE, and the possible mechanism of standardized mites allergens immunotherapy to allergic asthmatic children.Methods 31 children with mild to moderate allergic asthma were immunotherapy with standardized mites allergens for 25 weeks .The pulmonary function, the Der p specific IgG4,and the Der p specific IgE of patient were determined before treatment,25weeks later, all the proceedings would be determined again.Results The percentage of FEV1 and PEF to the prediction, PC20 , sIgG4 level and sIgE level were(97.87±23.44)%,( 99.95±29.52) % ,( 1.49±2.75 )mg/ml , 271.4 AU/ml ( 436.6 AU/ml) ,87.14 AU/ml(70.26 AU/ml) at the beginning, but they changed into(114.81±17.65)%,( 116.51±19.52) %,( 7.31±7.02) mg/ml,2450.5 AU/ml( 2771.95 AU/ml ), 100 AU/ml(59.25 AU/ml )after treatment. There is significantly increased after specific immunotherapy in each item (P<0.01) except sIgE (P>0.05).Conclusion The Standardized mites allergens immunotherapy made a good effect to the pulmonary function of allergic asthma children .Increased sIgG4 plays a great role of protection in specific immunotherapy.
