OBJECTIVEBaesd on the rat models with NAFLD feeded on high glucose, we studied the actions of ChREBP and its target genes ACC,FAS in the mechanism of NAFLD, by detection the expression levels of mRNA and protein in ChREBP,ACC and FAS ,and the bind conditions between ChREBP and carbohydrate response element (ChRE) of ACC and FAS promoters at different time point in rat liver.METHOD32 Sprague-Dawley(SD) rats were divided randomly into high glucose group and control group, and were sacrificed at the end of 4th, 8th and 12th weeks respectively. The level of ALT, AST, TC and TG in blood collected from back abdominal aorta after 12 hours'fasting were measured. Adipose degeneration of liver was observed by HE staining. Expressions of ChREBP in both mRNA and protein levels at different time point were detected using RT-PCR and Western blot respectively. The bind conditions between ChREBP and carbohydrate response element (ChRE) of ACC and FAS promoters at different time point were analyzed with chromatin immunoprecipitation assay(CHIP).RESULTSRat models with NAFLD feeded on high glucose dietsweresuccessfully prepared. The level of ALT, AST, TC and TG increased with time extending (P<0.01). Adipose degeneration of liver increased, and the expression of ChREBP, ACC and Fas in both protein and mRNA levels increased as compared to the control group (P<0.01). Expression of ChRENA also increased(P<0.01).CONCLUSIONThe expression of ChREBP were increased by high glucose diets, which participate in the formation of nonalcoholic fatty liver through up-regulation ACC and FAS.
