OBJECTIVE: 1. To prepare and characterize curcumin solid lipid nanoparticles(SLN-Cur) with stearic acid as carrier, using modified microemulsion method. 2. Investigate the effect of SLN-Cur alone or in combination with cisplatin (DDP) on the growth of human ovarian cell line SKOV3 and apoptosis induction.METHODS: 1. Curcumin solid lipid nanoparticles (SLN-Cur) were prepared by modified microemulsion method; Use transmission electromicroscope to study the shape of the particles; Use particles characterization and zeta potential analyzer to study the diameter and zeta potential of SLN-Cur in suspension; Use UV-VIS Spectrophotomery to determin the entrapment efficiency. 2. SKOV3 cells were respectively treated with Cur, DDP, SLN-Cur, DDP+ Cur and DDP+ SLN-Cur. Observe cell morphology variation and cell ultrastructure; Detect the apoptosis rate of SKOV3 cell and the expression of apoptisis-associated gene proteins.RESULTS: 1. The SLN-Cur prepared by modified microemulsion was spherical and the zeta potential were 341.8 nm and–38.7 mV, respectively, and the entrapment rate was 95.1%; After 60 days, the entrapment efficiency was almost unchanged. 2. SLN-Cur could effectively inhibit SKOV3 in a dose and time dependent manner; Simultaneous administration of SLN-Cur and DDP can result in synergistic cytotoxicity and apoptosis for SKOV3 cells; The expression of Bcl-2 was down-regulated and the expression of Bax was up-regulated.CONCLUSION: 1. The entrapment ratio of SLN-Cur and the stability of SLN-Cur are high and good. The modified micromulsion for SLN-Cur preparation is feasible in future research and application. 2. The rate of cell proliferation and the rate of SKOV3 cell line apoptosis by SLN-Cur alone or combined with DDP was significantly higher.
