Objective:To investigate the effect of serum on THP-1 cell TLR4/NF-κB signaling pathway in obese people with various metabolic syndrome components.Methods:We observed TLR4 mRNA and protein expression onTHP-1 cell, the level of NF-κB p65 phosphorylation in THP-1 cell and IL-1βand TNF-αexpression in culture supernatant. THP-1 cell was cultured in serum from volunteers with various metabolic syndrome components for 24h and 48h.THP-1 cell line was cultured in culture media with 15% serum of 50 volunteers divided into 5 groups of 10 each, with various degrees of metabolic syndrome components (healthy controls, simple obesity, obesity with hyperglycemia, obesity with hyperlipidemia, obesity with hyperglycemia complicated by hypertension and hyperlipidemia). TLR4 mRNA was measured by RT-PCR,TLR4 protein content and the level of NF-κB p65 phosphorylation were measured by western blotting,the expression of IL-1(3 and TNF-αwere detected by ELISA.Results:The TLR4 mRNA and protein content, the level of NF-κB p65 phosphorylation in THP-1 cell line, IL-1βand TNF-a expression in culture supernatant cultured in the 5 groups of serum were statistically significant. The group of serum from volunteers with obesity with hyperglycemia complicated by hypertension and hyperlipidemia, had the highest, followed by obesity with hyperglycemia, obesity with hyperglycemia, simple obesity and healthy controls respectively(P<0.05). The TLR4 mRNA and protein content, the level of NF-κB p65 phosphorylation in THP-1 cell line (P<0.05), and the IL-1βand TNF-αexpression in culture supernatant cultured in the serum group of obesity with three metabolic syndrome components were time-dependent.Conclusions:Serum in simple obese people and obese people with various metabolic syndrome components can induce TLR4/NF-κB signaling pathway on THP-1 cell then excrete inflammation factor.The more the metabolic syndrome components, the higher the levels in TLR4/NF-κB signaling pathway activation and ability of inducing inflammation.
