Objective:to explore the differences of chemical composition on drinking water organic extractions by the (XAD-2) macroporous resin and the Waters company Oasis HLB columella,and to compare reproductive toxicity of the two organic extractions in male mice. Methods:through XAD-2 macroporous resin and Waters company Oasis HLB columella respectively to extract a drinking water extracts, then using gc-ms to detect and analysis its composition,Through in vitro single-celled gel electrophoresis experiment and in vivo animal experiments, to analyze the two approaches extraction of organic extracts, between the two approaches, and between two kinds of methods with negative control group, the mice testicular tissue in testicular tissue malondialdehyde (MDA), superoxide dismutase (SOD), acidic phosphatase (ACP), alkaline phosphatase (AKP), gamma-glutamyl GGT(y-GT) level,serum male hormone testosterone(T)and follicle-stimulating hormone (FSH) levels if there is a certain difference in (1.25 L water organic extracts/kg) concentrations, middle dose group (2.5 L water organic extracts/kg), (5.01 water organic extracts/kg) concentrations. Results:XAD-2 macroporous resin compared with Waters company Oasis HLB solid-phase extraction columella drinking water have more organic types. Among them for phthalic acid ester substances, Waters company Oasis HLB solid-phase columella compared with XAD-2 macroporous resin,concentration efficiency increased as molecular weight gradually heighten. For in vitro single-celled gel electrophoresis experiment, of two methods of concentrated organic matter in low concentration (100ml raw water/ml broth) compared to negative nutrient-containing medium, there was no difference of the influence of generative cell. in the high-dosage (400 ml raw water/ml broth), compared with the control group and the solvent P value were less than 0.05. there are no differences between between XAD-2 macroporous resin in dose group of (200ml raw water/ml broth) and Oasis HLB columella in dose group of (200ml raw water/ml broth), and between XAD-2 high-dosage groups (400ml raw water/ml broth) and Oasis HLB high-dosage groups (400mlraw water/ml broth) also have no differences. By in vivo animal study found that organic extracts of drinking water through the metabolism of mice, the determination of the indicators show its effect is not obvious reproductive toxicity in mice. Conclusion:XAD-2 macroporous resin and Waters company Oasis HLB solid-phase extraction in drinking water columella organic extracts, there is a certain difference, but for phthalic acid ester substances, Waters companyOasis HLB compared with XAD-2 macroporous resin, its extraction efficiency increased with molecular weight increased, but two extraction methods of extraction of net end water on male mice, the influence of the reproductive system is not obvious, its may be its composition is complex and diverse.
