Purpose:To prepare the serial sections of cat optic nerve, search for an exact method of staining and registration, and establish a visible virtual cat optic nerve model by the three-dimensional reconstruction program.Methods:Based on the paraffin section method. The serial sections images were obtained by using Motic microscope and Motic software and saved as the format of JPEG. Conditioning the images and registration them by handwork. In the end, the three-dimensional model of cat optic nerve was reconstructed in computer by Amira 3.1.1 from the two-dimensional images.Results:After staining, the perineurium can be separate from other tissue in most sections. The virtual cat optic never was succeeded reconstruction by using Amira 3.1.1. And the reconstructed cat optic nerve can be freely rotated and incised and restored.Conclusions:Our study suggests that the method of without consult of registration in the main, and the method of 3D models reconstructed from serial histological sections of optic nerve is feasible.
