ObjectiveTo observe the effect of MK801 on the expression of Bcl-2,Bax and C-fos gene and its protective effects to the nerves in the hippocampus CA1 region of the rats after the brain ischemic-reperfussion injury.Methods212 male SD rats were randomly divided into 3 groups: normal group, ischemia-reperfusion group and treatment group.1.the normal group,total number 64,normally feeded,use the HE staining,immunohistochemical ,Western-blot methods to observe the morphology, the expression of the Bcl-2,Bax,C-fos gene in the hippocampus.2.the ischemic-reperfusion group and the treatment group ,74 rats respectively,both of the two group choose 64 rats and were given the cerebral ischemic–reperfusion treatment,And the treatment group were given the MK801 (0.5mg/Kg) at the same time interval,all of the rats were executed after the ischemic–reperfusion of 2h,6h,24h and 48h.Then use the HE staining, immunohistochemical,Western-blot methods to observe the morphology, the expression of the Bcl-2,Bax,C-fos gene in the hippocampus.the other 20 rats in the two groups were executed after the reperfusion 6h and 48h and observe the total area of ischemic.ResultsContrast to the normal group, the expression of the Bax and the C-fos gene was significantly increased in the model group and the MK801 group in every time interval, the difference has the statistic significance(P<0.05),and the expression of the Bcl-2 was significantly decreased in every time interval,the difference has the statistic significance(P<0.05); Contrast to the model group, the expression of the Bax and the C-fos gene was significantly decreased in the MK801 group ,the difference has the statistic significance(P<0.05), and the expression of the Bcl-2 was significantly increased in every time interval.Contrast to the model group,the total area of infarction of the cerebral in the MK801 group were significantly decreased than in the model group,the difference has the statistic significance(P<0.05).ConclusionsMK801 could inhibit the apoptosis progress by refrain the excitatory amino acid receptors and has the protective effect to the neurons in the hippocampus CA1region.MK801 could improve the expression of the Bcl-2,inhibit the expression of the Bax, and increase the expression of C-fos, by this way MK801 could inhibit the apoptosis progress. 6h and 48h after the ischemic-reperfusion are the damage peak time, in these two time intervals ,the neurons apoptosis are more severe than the other time intervals. It has provided a better time window reference for the preventive and curative ischemic-reperfusion injury in clinic .
