ObjectiveCytokine-induced killer cells (CIK) have powerful cytotoxic activity which can kill cells and dendritic cells (DC) are the most powerful antigen-presenting cells. This paper aims to study the immuno-effect of cultivating DC-CIK on killing leukemia. To provide the basis for blood of cancer patients'immunotherapy through the mechanism research of DC - CIK cells in vitro.Methods1. Peripheral blood of normal persons was processed into the anti- coagulation and the isolation of peripheral blood mononuclear cells using lymphocyte separating medium. PBMNC were induced to CIK, DC and DC-CIK. Then the proliferative ability of cells was analyzed through analyzing growth curve, and immunophenotype of cells was measured by flow cytometry.2. K562 served as target cells, and PBMNC, CIK, DC, DC-CIK as effector cells. The killing activity was assayed by MTT. The ratios of effector to target for every group were 10:1and 20:1.3. Production of IL-17 by PBMNC, DC, CIK and DC-CIK was detected by ELISA.Results1. Highly purified DC and CIK cells were induced by PBMNC. They highly showed special immunophenotype of cells by flow cytometry.2. With the increased ratio of effector to target, effector killing activity of each group also increased; for the same ratio, the killing activity of each group was different, of which DC-CIK was the strongest(P<0.05).3. The amount of secretion of cytokine IL-17 for every group were(4.15±0.77)ng/L,(4.56±0.39)ng/L, (4.95±0.51) ng/L and(8.36±0.39)ng/L. In DC-CIK group, IL-17 was more than those of any other group(P<0.05).Conclusions1. It showed that DC-CIK has a stronger anti-tumor activity, and the toxicity of DC-CIK against leukemia cells was significantly higher than simple application of CIK or DC cells.2. DC-CIK against leukemia cells was related to the immunological effect of highly secreted cytokine.
