Objective To determine the expression differences of Fibulin-2 gene in breast cancer tissueand corresponding adjacent normal breast tissue as well as breast fibroadenoma tissue andcorresponding adjacent normal breast tissue by FQ RT-PCR and Western blot techniques, for theevaluation of the impact on the invasion, metastasis and progress of breast cancer and its clinicalsignificance .Methods This study has collected 30 cases of specimen of breast cancer and correspondingadjacent normal breast tissue (breast cancer group), 32 cases of specimen of breast fibroadenomaand corresponding adjacent normal breast tissues (breast fibroadenoma group). After sending tothe general pathological examination specimens were immediately played on ice and thenquickly transferred to -80℃refrigerator for the performance of FQ RT-PCR for detectingFibulin-2 gene expression and Western blot for analyzing Fibulin-2 protein .Results⑴Breast cancer group: The expression of Fibulin-2 mRNA was average 6.149-foldlower in breast cancer tissues than in adjacent breast cancer tissues (P<0.05), among this 10-foldlower in 15 breast cancer tissues, 4-fold lower in 8 breast cancer tissues and 2-fold lower in 3breast cancer tissues .⑵Breast fibroadenoma group: There was no significant difference in theexpression of Fibulin-2 mRNA in fibroadenoma of the breast and the adjacent normal tissue(P>0.05) .⑶Furthermore, Western Blot analysis showed that there was also significantdifference in Fibulin-2 protein expression between the breast cancer and adjacent tissues, which was in accordant with the results of FQ RT-PCR analysis of Fibulin-2 mRNA levels .Conclusion The expression of Fibulin-2 mRNA and protein in breast cancer wassignificantly reduced compared with adjacent normal breast tissue, and it has related with thedegree of tumor differentiation, so it is suggesting that Fibulin-2 may be associated with theprogression, metastasis and infiltrating potential of breast cancer. Fibulin-2 can be used asindicators of molecular biology malignancy of breast cancer .
