Objective: To explore the appropriate dose of oxaliplatin in retrograde isolated hepatic perfusion through observering of the effect of oxaliplatin to rabbits with VX-2 liver tumor by retrograded isolated hepatic perfusion. Methods: New Zealand white rabbits were randomly divided into three groups, The retrograded isolated hepatic perfusion model was established after the success of VX-2 liver tumor model. Group A (n=10) and group B (n=10) were infused oxaliplatin doses of which were 12mg/kg and 16mg/kg respectively via hepatic artery for 5 min, group C(n=10) was 5% glucose. Outflow and peripheral venous blood after perfusion immediately and venous blood after 24 hours were collected. Also the tumor and liver biopsy specimens of three groups were taken after 24 hours. ALT, AST, and the concentration of oxaliplatin in blood and liver tumor tissue samples were detected. The tumor angiogenesis growth factor (VEGF) were detected by immunohistochemistry and tumor specimens structural changes by electron microscopy and pathology.Results: ALT and AST in three groups were increased compared with the preoperation group, The most obvious change was B group, Outflow was significantly higher than peripheral venous in A group and B group. Cells were changed in A group and B group though VEGF, pathology, electron microscopy , The most obvious change was B group. Conclusion: Oxaliplatin can damage and inhibition the VX-2 liver tumor obviously. However, when the dose was increased to a certain amount it would be generate injury on the liver. The dose of oxaliplatin can apply to RIHP is at least 12mg/kg.
