Objective:To observe the effects of berberine on glycolipids metabolism in rats with insulin resistance induced by high fructose and fat diet, and investigate the expression of ERS markers in liver, fat, muscle tissue to reveal the therapeutic mechanism of berberine on Diabetes and Metabolic Syndrome.Methods:The rats with insulin resistance were induced by high fructose and fat diet for 8 weeks. Then the rats were randomly divided into 3 groups:model group, Berberine group and PBA group. Meanwhile, a normal group fed by regular diet was also designated. Collect the serum and organs from the rats 6 weeks after treatment , and study the effects of Berberine on blood(FBG GTT), blood lipids (TCH, TG, HDL-C, LDL-C) and the expression of molecular chaperones and marker molecule related to ER stress(ORP150, PERK, eIF2α) with experimental techniques such as biochemistry, radio immunoassay, and western blot. Results:Compared with the model group, after 6 weeks'treatment with Berberine, the result of GTT was improved significantly. The level of body weight and FBG were decreased. The level of TCH, TG, LDL-C were decreased while HDL-C and ISI were enhanced. The expression of ORP150 was down-regulated and the phosphorylation of PERK-eIF2αwas suppressed.Conclusion:Rats model with insulin resistance could be induced by Feeding with high-fructose and high-fat diet. Berberine could prevent fructose-induced insulin resistance in rats, and improve its glycolipids metabolism. The therapeutic mechanism of berberine on insulin resistance might be related with its regulation on the expression of ORP150 and the inhibition on the phosphorylation of PERK-eIF2α, accordingly improved the ER stress.
