Objective To investigate the effect of grape seed proanthocyanidins extract (GSPE) on proliferation and migration of Tenon′s capsule fibroblasts of human. Methods TFb were treated with GSPE at different concentrations for 24h. The cellular viability was analyzed through MTT assay , wound healing model was used to detect the migration of TFb, immunohistochemistry staining method was used to observe the expression of p21. Results With GSPE treatment varying from concentrations of 10ug/ml to 40ug/ml , the proliferation of TFb decreased in a dose dependent manner (P<0.01), the number of migrated TFb were 46.80±11.35,29.60±7.20,10.80±5.50 per field (P <0.01) , decresded in a dose dependent manner compared with 86.00±11.47 of the control group(P <0.01).The results of immunohistochemistry demonstrate that the expression of p21 protein increased in GSPE-treated group, compared with the control group,the difference was statistically significant. Conclusion GSPE could inhibit the migration of TFb , and possibly through up-regulating the expression of p21 protein in inhibiting cell proliferation, These findings suggest that GSPE may be a good candidate for blocking development of filtering bleb scarring.
