In recent years, the transdermal drug delivery systems (TDDS) has been developed rapidly in pharmaceutical science. The uttermost advantage of TDDS is to reduce adverse side effects, to enhance therapy index and for the convenience of clinical usage. This research was focused on the study of hydrogel preparation of consituents extracted from Spina Gleditsiae, including the study of in-vitro antimicrobial activity and effective constituents,the study of toxicology,prescription study and the study of pharmacodynamics. The concrete job are as follows:1. A patented gasification and fulmination equipment was used to prepare the extracts of Spina Gleditsiae, whose diameter is from 50 nm to 100 nm. Also, extracts from Spina Gleditsiae with water and alcohol were prepared respectively. Antimicrobial activity of the constituents extracted with gasification and fulmination method is more significant than the other two, in which the constituents were obviously different. On the basis of some chemical tests, Folin-Ciocalteu method was employed for total phenol contents determination.The analysis method was proved to be precise,simple and reliable.2. To provide the basic datas for presicription study, dermal sensitization test,skin irritation test and acute toxicity test of extracts made by gasification and fulmination method were employed. And the results showed that the extracts were safe. 3. The influences of different main polymer, crosslinking agent, crosslinking regulator, adhesions and humectant on the hydrogel patch were studied by using peeling strength,gel strength and elasiticity as the assessing indexes. NP-700,Aluminium glycinate,tartaric acid,CMC-Na and glycerin were chose as a whole ingredient.Single factor investigation was used to determine the suitable contents of different adjuvants.The formulation was optimized by using orthogonal design with tack and peel as the indexes. Then, the optimization of formulatio with extracs were made in the end. The intial quality evaluation of the prepared hydrogel patch was carried out. And the results were meet the requirement according with Pharacopoeia of the People's Republic of China of 2005 edition.4. The patch would be used to study the skin irritation. As a result, the rabbit skin showed no any irritative and allergie response, which meant that the patch is safe for topical administration. Also, a skin abscess model in mice by infection of staphylococcus aureus was established to evaluate the pharmacodynamics of hydrogel patch. The results showed that the volume of skin abscess was higher in the infected mice than that in the drug-treated animals and the patch can inhibit the expansion of skin infection.In this dissertation, it was proved that hydrogel patch is a new drug system with great research value and potential, it can be used in surgery wounds,cosmetology and health care,normal cuts and skin infection popularly.
