Objective To study the relationship of radiosensitivity and expression of ATM and DNA-PKcs in human cervical carcinoma.Methods Immunohistochemistry was used to detect the expression of ATM and DNA-PKcs for 50 patients with cervical carcinoma and 20 patients with normal cervical tissue.Dased on the current radiotherapy,the patients of cervical carcinoma were divided into two groups,which were high radiosensitive and low radiosensitive .The expression of the two proteins,ATM and DNA-PKcs,were campared, and combined with clinical data, were analized the relationship with patients'age, clinical stage and differentiation.Results (1)The positive percentages of ATM in cervical carcinoma and normal cervical tissue were 22% and 60% respectively, the differences had statistical significance(P<0.05).(2)The positive percentages of DNA-PKcs in cervical carcinoma and normal cervical tissue were 62% and 40% respectively, the differences had no statistical significance (P>0.05).(3)The expression of DNA-PKcs in cervical carcinoma was higher than ATM, the difference had statistical significance (P<0.05).(4) In 50 cases of cervical carcinoma after radiotherapy, CR is 48% (24/50), PR is 52% (26/50).(5)The expression of ATM and DNA-PKcs in high radiosensitive group and low radiosensitive group were 8.3% and 34.6%,45.8% and 76.9% respectively, the differences of both had statistical significance (P<0.05).(6) Spearman correlation analysis showed that the expression of ATM and DNA-PKcs in cervical carcinoma had no correlation(r=0.251 P=0.078).(7)The differences of ATM and DNA-PKcs between different cervical carcinoma patients'ages, clinical stages and differentiations had no statistical significance (P>0.05).Conclusion (1)The expression of ATM in cervical carcinoma was lower than that in normal cervical tissue, ATM may take an important role in the progression of cervical carcinoma.(2)The high expression of ATM and DNA-PKcs lead to radioresistance,on the opposite,the low expression of them lead to radiosensitivity, ATM and DNA-PKcs may forecast radiosensitivity of cervical carcinoma.(3)The expression of ATM and DNA-PKcs had no obvious correlation in cervical carcinoma.(4)The expression of ATM and DNA-PKcs had no correlation with patients'ages, clinical stages and differentiations.
