In recent years, there were more and more reports about lead pollution and lead hazards to human beings (especially to children). Lead is persistent pollutant, and can not be disintegrated by biological metabolism in the natural environment. Lead is harmful to most of the systems in human, especially to bone marrow hematopoietic system and nervous system and kidneys. Lead can be absorbed into the body and some can be excreted through feces and perspiration, while the rest can be dissolved into the blood in a few hours and impede the synthesis of blood.Serum protein is the main ingredient in the blood plasma, and it has the function of storage and carriage. Serum protein is widespread in the blood and can be easily purified, so it became one of the earliest proteins studied by scientists. The starting point of this topic is to discuss lead and serum protein combination mechanism from the molecular aspects, also to discuss effective methods for reducing lead levels in the complex.First of all, this study studied the fluorescence spectrum of hybrid system constituting of lead and bovine serum albumin (BSA) in the buffer solution, and got the following conclusions: The extinction concerning Pb2+ for BSA is primarily static extinction, in other words, lead and BSA generated complex in the buffer solution.Secondly, this study used traditional dialysis method to reduce the lead levels in Pb- BSA complex. In the experiment, the mixture made by Pb(NO3)2 and BSA was used as poisoning simulation blood. The simulative dialysis device set up by ourselves was used to reduce the lead levels in poisoning simulation blood. The lead levels in the simulative blood were detected through the period of dialysis. Test results showed that, the lead levels in the acidic condition of poisoning simulation blood descended rapidly through dialysis, finally the concentration of lead was very low; while the lead levels in the neutral condition of poisoning simulation blood descended slowly through dialysis, finally the stable concentration of lead was rather high. In conclusion, traditional dialysis method can't be used to reduce the lead levels in Pb- BSA complex.Additionally, the dialysis device designed by ourselves was used to dialyze the poisoning simulation blood under neutral condition, while the lead ions not forming complex and other impurities were removed through dialysis, then the poisoning simulation blood was electrolyzed by the device designed by ourselves, after that the simulative blood was again dialyzed. In the meantime, the lead levels were detected in all the period. Test results showed that, electrolysis can be effective to reduce the lead levels in poisoning simulation blood and the lead generated by dialysis can be deposited on the electrodes.In conclusion, it can be inferred that: Pb(NO3)2 and BSA generated plenty of complex under physiological condition; traditional dialysis method can not be used to reduce the lead levels in Pb- BSA complex; electrolysis can be effective to reduce the lead levels in poisoning simulation blood, electrolysis is a kind of effective method to remove lead.
