Peels of Citrus reticulata being dried for more than three years becomes to be the Tangerine peels, and it was used as seasoning and preserved fruits. Abundant polysaccharide were found except Volatile oil and flavone in the peels. In this paper, modern isolation and analysis methods were used to do the research on the extraction, purification, characteristics, quantitative analysis of component monosaccharides, structure and bioactivities of citrus polysaccharide. The research offers some scientific basis and comprehensive utilization.1. Crude polysaccharide was extracted by acid water, and the best extraction was optimized with the orthogonal design. The result showed that the optimum condition for extraction process as follow: the amount of acid water(pH 4) added was 30 times that of dry-material, the temperature was 90℃and the total time of extraction was 90min. The yield and ability of anti-oxidant(FRAP) were 18.9% and 48.9 respectively. The citrus polysaccharide was classified as high-methoxly pectin with high expansibility and water-holding capacity.2. The decoloration effect of macroporous resins and ion exchange resins using static test of decoloration were studied. The ion exchange resins of D-201 was better that other resins and the optimum decoloration condition were with 8%(m/V) of resin, 75r/min of rotation speed and being extracted with Sevag 69times, the protein was deleted.3. A new HPLV-UV technique for the separation and analysis of 8 monosaccharides using 1-pheny-3-methyl-5-pyrazolone(PMP) as the labeling molecule of the reductive monosaccharides has been established by hing performance liquid chromatography-UV and C18 column. The acid-hydrolysis condition, derived condition and chromatography separation condition was optimized, and the optimized conditions are as follow:hydrolysis with 2M TFA in the temperature of 120℃reaction time is 40min, incubation temperature is 70℃, ratio of CH3Cl and monosaccharide is 1:1 of 3 times extraction.4. CP was separated by ion exchange chromatography DEAE-Sepharose Fast Flow and 4 fractions were gained. Three of them were purified by gel permeation chromatography G-100 and CPA-Ⅰ,CPA-Ⅱ,CPA-Ⅲwere gained. They were analyzed by TSK G-5000 PWxL and TSK G-3000 PWxL collumn HPLC. The result showed that CPA-Ⅰ,CPA-Ⅱ,CPA-Ⅲhave good purity, molecular weight were 8.0×104 Da, 2.6×105Da, 2.4×105Da respectively. IR, NMR spectra indicated that the polysaccharide was mainly composed of (1→4)-linkedα-GalpA, some of the galacturonic acid were methyl esterification and mannose, ribose, rhamnose existed as pyranoid in the side chain or at the end group.The result showed CPN consisted of mannose, rhamnose, galacturonic acid, glucose, galactose and xylose and arabinose in the molar contents of 0.34, 0.07, 5.30, 1, 4.71, 0.61, 14.7 respectively; CPA-Ⅰconsisted of mannose, ribose, rhamnose, galacturonic acid, glucose, galactose and arabinose in the molar contents of 0.94, 0.05, 1.33, 56.65, 1, 7.04, 19.38 respectively; CPA-Ⅲconsisted of mannose, rhamnose, galacturonic acid, glucose, galactose and arabinose in the molar contents of 2.02, 1.98, 122.66, 1, 8.85, 26.26 respectively.5. An experimental model of oxidative damage induced by hydrogen peroxide was established based on the optimal concentrateon of H2O2. The results showed that the optimal concentration determined was 0.1875mmol/L. The citrus polysaccharide has remarkable antioxidative bioactivities and the concentration between 0.078125 and 2.5 mg/mL could enhance the antioxidantive bioactivities...
