Objectives:Researchers detects that Cyclooxygenase-2(COX-2) express higher in most tumors, and the level of COX-2 parallel to the degree of metastasis and the TNM stage. We adopted cell lines with diverse differentiation, and investigated the relationship between the level of COX-2 and the degree of differentiation, at both protein and genes levels, prepared for later experiment.Methods:We employ Western blot and Real time quantitative RT-PCR to measure the level of COX-2 in gastric cancer cell lines AGS, SGC-7901, BGC-823, MGC-803 etc.Results:At the level of protein:SGC-7901 express the highest of COX-2 than the others, second is AGS, and third is BGC-823, MGC-803 do not express COX-2. At the level of genes:AGS express the most of COX-2, next is SGC-7901, third is BGC-823, MGC-803 is least.Conclusion:The expression of COX-2 is a common phenomenon in gastric cancer cell lines and it is closely related to differentiate of tumor cells. Objective:To study the relationship of COX-2, NF-κB, Snail and E-cadherin in human gastric carcinoma cell line of BGC-823 which expressed the least COX-2, and probe the possible molecular mechanism of COX-2 regulates the expression of E-cadherin.Methods:After treat with the different concentrates of celecoxib and PGE2 at different time points on human gastric carcinoma cell lines of BGC-823, real time PCR analysis the mRNA expression of COX-2,NF-κB,Snail and E-cadherin.Results:The COX-2 metabolites of PGE2 can up-regulate expression of NF-κB and Snail and down regulate expression of E-cadherin, accompany by different concentration and time of PGE2 at genes level. The level of COX-2, NF-κB, Snail and E-cadherin is not parallel with the concentration and tine by celecoxib.Conclusion:COX-2 promotes invasion in gastric carcinoma, which carry out by Snail directly or activating NF-κB indirectly regulate on E-cadherin.
