Objective To evaluate the targeted therapeutic efficiency of 131I-Herceptin on human ovarian carcinoma with high expression of HER-2/neu in vivo and in vitro.Methods There were five groups in the experiment, including 131I-Herceptin plus DDP gr, 131I-Herceptin gr, Herceptin gr, DDP gr, NS gr. The Herceptin was combined with 131I by Iodogen method. The inhibitory effect of each experimental group on human ovarian carcinoma cell line SKOV-3 in each experimental group was explored by MTT method. The tumor-bearing nude mice were randomly divided into five groups. Drug was injected into the tail vein of the nude mice once a week for six weeks. The size of the tumor was measured weekly and the mice were killed a week after last treatment. The inhibitory ratio of tumor growth was calculated and tumor tissues were observed by using HE staining. The apoptotic index was analyzed by using Tunel technique. The P53 and HER-2 expression in xenograft tumors were analyzed by using immunohistochemical staining.Results The 131I-Herceptin inhibited the growth of SKOV-3 cells significantly, and there was strong synergistic effect between 131I-Herceptin and DDP. 131I-Herceptin plus DDP group have higher apoptotic index than any other experimental groups, its expression rates of P53 and HER-2 were less than the others.Conclutions 131I-Herceptin could inhibit the growth of human ovarian carcinoma with high expression of HER-2/neu in vivo and vitro, the effect was associated with the expressions of HER-2 and p53. Its therapeutic efficiency could be enhanced by using with DDP.
