Objective To analysis the sequence characteristics and the genetic polymorphism of two Y-STR loci DYS510 and DYS542,then discuss the possibility of applying them to forensic science by investigating 135 unrelated Han males of the two Y-STR loci in Shanxi population, to meet the need of more genetic markers in forensic practice and genetic analysis.Methods To collect respectively 135 unrelated males'venous blood 500μl, who live in Shanxi province. Different tissues of one corpse were analyzed including blood, muscle, heart, liver et al.36 father-offspring pairs were investigated. The blood of 25 females were collected.All blood-specimens were anticoagulanted with EDTA. The DNA were extracted using the method of phenol-chloroform after ingested by proteinase K, choosed two Y-STR loci DYS510 and DYS542 from GDB, and amplified with PCR., the products were analysised by agarose gel electrophoresis and 8% non-denaturing polyacrylamide gel electrophoresis and associated with silver stained.Results There are total 7 alleles in DYS510 site in Shanxi Han population, called DYS510*14,DYS510*15,DYS510*16,DYS510*17,DYS510*18,DYS510*19,DYS510*20, respectively with the allele frequencies of 0.0222,0.0519,0.0297,0.3259, 0.4000,0.1481,0.0222,the discriminating power,gene dinersity, possibility oa exclution were 0.7126.There are total 5 alleles in DYS542 site were found, called DYS542*14,DYS542*15,DYS542*16,DYS542*17,DYS542*18 respectively with the allele frequencies of 0.1037,0.4000,0.3556,0.1259,0.0148,and the gene diversity was 0.6918,the discriminating power and excluding probability of paternity were the same. There are not amplified products among females. The typing results of different tissues from one individual were identical. The amplified products of female's samples were negative. Mutations were not found from 36 father-offspring pairs.Conclusion These results indicate that the DYS510 and DYS542 loci are high genetic polymorphic, the haplotype diversity shows good discriminating power and high excluding chance of paternity.They are to be useful genetic markers in forensic science and human evolution studies.
