Objective:To investigate the expression of aquaporin 1(AQP1) and the effect of hydroxyethyl starch 130/0.4 in renal damage with severe acute pancreatitis(SAP) rats , and explore the correlation between the expression of aquaporin 1 and renal damage with SAP.Methods:50 male SD rats were divided randomly into six groups:sham operation group(SO group);severe acute pancreatitis group(SAP group) and hydroxyethyl starch 130/0.4 treatment group(HES group),every group divided into 6h,24h two time points,SAP group 24h 10 rats,the remaining time point 8 rats.The SAP model was constructed by injecting 5% sodium taurocholate into the pancreatic-biliary duct , Femoral vein administration after operation.Serum amylase(AMY),urea nitrogen(BUN),creatinine(Cr) and histological change of the pancreas and kidneys were measured every time point,Serum tumor necrosis factor alpha(TNF-α) was detected by enzyme linked immunosorbent assay(ELISA),The expression of aquaporin 1(AQP1) mRNA in kidney tissue was analyzed by reverse transcription-polymerase chain reaction(RT-PCR),The kindey tissue was detected by immunohistochemical for AQP1 expression.Results:As compared with the SO group,Serum TNF-αlevel in SAP group increased significantly after SAP onset,AQP1 mRNA expression in the kidney down-regulated,The expression of AQP1 decreased,Serum AMY,BUN,Cr levels elevated 6h and 24h after SAP onset respectively(P<0.05).As compared with the SAP group,In HES group,Serum TNF-αlevels decreased markedly 6h[(97.14±6.97)pg/ml vs. (78.29±4.77)pg/ml , t=-6.317 ,P<0.01],24h[(148.49±24.77)pg/ml vs. (112.50±7.11)pg/ml,t=3.951, P<0.01] ; BUN levels decreased markedly 6h[(23.43±2.27)mmol/L vs. (16.93±1.48)mmol/L,t=-6.768,P<0.01],24h[(38.21±5.47)mmol/L vs. (27.61±1.95)mmol/L,t=-5.165,P<0.01];Cr levels decreased markedly 6h [(63.63±10.08)umol/L vs. (45.13±7.55)umol/L,t=-4.154,P<0.01],24h [(123.88±11.74)umol/L vs. (75.75±5.28)umol/L,t=-10.572,P<0.01];AQP1 mRNA expression was up-regulated 6h[(0.395±0.027) vs. (0.662±0.027),t=19.976,P<0.01],24h[(0.299±0.016) vs. (0.531±0.013),t=31.999,P<0.01];AQP1 expression was increased 6h[(64970.97±21203.05) vs. (132176.50±19406.66),t=6.613,P<0.01],24h[(65268.66±21258.59) vs. (130403.80±15604.28),t=6.986,P<0.01].The pathological change of the kidneys and pancreas attenuated as compared with SAP group.Conclusions:The down regulation of AQP1 in kindey is one of the causes of renal damage with SAP;Hydroxyethyl starch 130/0.4 could inhibit serum levels of TNF-α, up-regulate AQP1 mRNA expression , increase the expression of AQP1 and attenuate renal dysfunction with SAP.
