Objective:To study the inhibiting effect of ursolic acid(UA) on human breast carcinoma MCF-7 cells and whether its potential mec-hanism is associated with endoplasmic reticulum stress or not.Method:MCF-7 cell was cultured in vitro and treated by ursolic acid with a series of concentration (10μmol/L,20μmol/L,30μmol/ L,40μmol/L,50μmol/L and 60μmol/L), then detect the absorbance at 490nm by MTT assay,calculate the cell proliferation inhibition rate;The molecular mechanisms involved in the effects of UA on MCF-7 cells were studied by western blot,treated with a concentrati-on of 20μmol/L UA for 6 hour and 24 hour,detect the expression of CHOP/GADD153.Results:MTT assay showed that UA had a anti-proliferation e-ffect on MCF-7 cells in a dose-dependant manner.The inhibition rat-io elevated significantly compared with control group(p<0.01) after treated with Ursolic Acid of 10μmol/L for 24 hour,and correspo-nding rised for UA's concentration.Western blot didn't show signify-cant difference in expression of CHOP/GADD153 among the contr-ol group,cells treated with a concentration of 20μmol/L UA for 6 hour and a concentration of 20μmol/L UA for 24 hour(P>0.05).Conclusion:UA shows significant inhibiting effect on MCF-7 cells in a dose-dependant manner, but the mechanisim isn't relevant to endoplasmic reticulum stress,or didn't take a dominant place.
