| Objective:Our research used three strains of Lactobacillus separated from normal intestine of human, to investigate the effect on weight, blood lipid and obesity in rats fed with high-fat diet. Then we research the change of Intestinal flora's composition and distribution in obesity rats, also these Lactobacillus'effect on the composition and distribution of intestinal flora. It was aimed to detect the different effect of Lactobacillus strains on obesity, analysis the dynamic change of intestinal floras though obesity,and the different bacterial strain's effect in different place in bowel. So we can provide the based on for research the effect of intestinal flora on obesity and the research probiotic's function of lose weight and lower lipid.Methods:60 SD rats were randomly divided into 6 groups. Group K were normally fed, While the other five group were fed with high-fat diet and various Lactobacillus strains and Orlistat at the same time:Group A(strain M7-5), Group B(strain 08H16), Group F(strain 9-41-A), Group F positive drug (Orlistat), Group K model group (NS). We take their fresh stool measured intestinal floras analyses respectively in 0,2,4,6 week. After 6 weeks, all rats were executed. The effects of various Lactobacillus strains on rat fed with high-fat diet were determined with weight change, blood lipid, Lee's index. Microbial condition in large and small intestine contents were measured intestinal floras analyses. The formation and distribution of intestinal flora were determined with comparing the different weeks and location's intestinal floras analyses.Results:The body weights increased from 283.83±10.78(Group normal) to 348.71±18.01(Group model) in sixth week, it means we make model successfully. The body weights decreased significantly in all Lactobacillus strains groups. The weight decreased to 335.71±16.47 (Group A),331.88±12.60 (Group B),325.57±10.58 (Group F) respectively (P<0.05). Also the weight decreased to 331.96±9.20 in group drug (P<0.05). The Lee's score in group G were decreased in compared with the group model (P<0.05).TG level of group A,B & F were decreased obviously (P<0.05), and TCH level of all groups were decreased, group B&F were decreased obviously(P<0.05). In intestinal floras analyses, the Enterobacteria were increased obviously(P<0.05), the Bifidobacterium, the Bacteroides and B/E scores were decreased obviously(P<0.05), the Enterococcus and the Lactobacillus were decreased slightly(P>0.05) in group model. In group A the Bifidobacterium, the Lactobacillus and the Bacteroides were increased (P>0.05). In group B&F the Lactobacillus were increased (P>0.05), the Enterobacteria were decreased obviously (P<0.05).Conclusion:1. Lactobacillus M7-5 strain,08H16 strain and 9-41-A strain have reduced body weight and decreased the blood-lipid level definitely. The 08H16 strain has the best effect, even close to Orlistat.2. Lactobacillus M7-5 strain can increase the Bifidobacterium, the Lactobacillus and the Bacteroides. Lactobacillus 08H16 strain and 9-41-A strain can increase the Lactobacillus and the Bacteroides, and decrease the Enterobacteria obviously. They can improve the effect of obesity from different ways.3. The intestinal flora change obviously in obesity prophase.4. The Lactobacillus get though a persistent effect and act on small intestine. |
