Partâ… :Classification of Genus Malassezia and their distribution in common Malassezia-related DiseasesObjective:To study the classification of Genus Malassezia and their distribution in two common Malassezia-related skin diseases.Methods:Using standard strains as control, classified Malassezia strains from pityriasis versicolor and Malassezia folliculitis applying ITS2 region PCR and sequencing. And analyzed distribution of each species in these two diseases.Results:Isolated 6 species and 114 strains Malassezia in total, including 47 strains of Malassezia globosa (41%),40 strains of Malassezia sympodialis (35%),20 strains of Malassezia furfur(17.5%),4 strains of Malassezia obtusa (3.5%),2 strains of Malassezia yamatoensis(1.8%) and 1 strains of Malassezia dermatis (0.9%).The construction of species in the two diseases was associated with type of the diseases. Isolated Malassezia sympodialis from almost half of the patients with pityriasis versicolor and Malassezia globosa were isolated from most of the patients with Malassezia folliculitis.Conclusion:The construction of Malassezia species varied in different diseases and Malassezia globosa is associated with Malassezia folliculitis closely; PCR and sequencing of ITS2 region in rDNA can be applied to the classification and identification of Malassezia strains.Part II:Establishment of identification-database of Common Malassezia using Biolog Automatic Analyzer for MicrobesObjective:To establish identification-database of common Malassezia using Biolog Automatic Analyzer for Microbes and investigate prospect of this system applied.Methods:Malassezia were identified into a species level using phenotypic methods and PCR amplification and sequencing. Malassezia furfur, Malassezia sympodialis and Malassezia globosa were inoculated into FF microplates and the utilization of 95 different carbon sources were recorded. Growth and reaction spectrum of the strains were described and identification database was set up.Results:There are obvious differences of carbon utilization among different strains. And the utilization trends of the same species are almost identical.Conclusion:Biolog Automatic Analyzer for Microbes has the ability to identify this three common Malassezia into species level, so we can speculate the application prospect of this system in identifying different Malassezia species.
