Background:Assessment of lymph node status of gastric carcinoma is the key to options of reasonable treatment before surgery. The complete region lymph node dissection is of great importance to improving survival rate. TNM stage is based on the size, shape, position and enhancement degree of lymph node, but for small metastasis of lymph node, its size can be normal. The ability of lymphatic targeting and adsorbing of ACNP (activated carbon nanoparticles) can be used to identify lymph nodes with ACNP absorbing contrast agent.Part OneObjective:To observe the distribution and migration of ACNP (activated carbon nanoparticles) injected into rats'right hind legs and to provide the basis for the ability of lymphatic targeting. Methods:The SD rats were respectively injected into rats' right hind legs with ACNP. At 0.5,1,2,3,5,6,12h after drug administration, execute the rat and observe the dyeing of popliteal fossa and inguinal lymph node of each rat. Results:At 1h after drug administration, the rat's popliteal fossa lymph node turned into black, but the rat's inguinal lymph didn't change. At 2h after drug administration, both the rat's popliteal fossa and inguinal lymph node turned into black and the popliteal fossa lymph node'colour is deeper. Conclusion:The ACNP show the ability of lymphatic targeting and its migration is in the same direction as the flowing of lymph.Part TwoObjective:Make use of the ability of lymphatic targeting and adsorbing drugs of ACNP to observe and analysis MDCT (Multi-Detector Computed Tomography) image of popliteal fossa lymph node of SD rats after injection of ACNP absorbing CT contrast agent (Omnipaque). Methods:SD rats were divided into 3 groups randomly. ACNP absorbing Omnipaque, saline and Omnipaque were respectively injected into rat's right hind legs of each group. At 0.5,1,1.5,2,3,4,6h after drug administration, scanning the rats with Siemens SOMATOM Sensation 64-CT and analyzing the CT value of popliteal fossa lymph node of each rat. Results:The CT value of SD rat's popliteal fossa lymph node after injection of ACNP absorbing CT contrast agent (Omnipaque) arrived at the peak at 2 hour and gradually decreased, which is always higher than the CT value of rats after injection of only CT contrast agent. Conclusion:The ACNP absorbing CT contrast agent (Omnipaque) is a sustained-release drug. Animal experimental study shows the effect of ACNP absorbing CT contrast agent (Omnipaque) labeling the rats'lymph nodes on CT.Part ThreeObjective:Make use of the ability of lymphatic targeting and adsorbing drugs of ACNP to observe and analysis MDCT image of popliteal fossa lymph node of SD rats with lymph node metastasis after injection of ACNP absorbing CT contrast agent (Omnipaque).
