In recent years, much attention has been devoted to the nanovehicle self-assembled from amphiphilic copolymer, which is used for drug carrier. Despite the discovery and development of anticancer drugs for the treatment of cancers, their clinical outcomes have been disappointing due to severe side effects and poor solutions. So it's very important to develop novel biomaterials used for drug-carrier, especially smart drug-carrier. Although nanovehicle systems have been evaluated and shown to be partially successful for cancer treatment in various in vivo models and clinical settings, there is still room to further improve their therapeutic effects.In the first section, we synthesized a novel amphiphilic pH-sensitive copolymer poly(PDM-co-HEA), which could self-assemble into nanoparticles with sizes of about 167 nm (PDI = 0.03), and the sizes of nanoparticles increased from nano-scale to almost micro-scale in acidic environment, which was a very important property for drug release. Nile Red was employed as hydrophobic and fluorescence molecules to mimic the drug delivery and release. The result showed that nile red could be delivered into the Hep3B cells by the nanoparticles and released in cytoplasm determined by fluorescence microscopy.In the second section, we describe the synthesis of well-defined (PCL)2-PtBA star block copolymers using a novel heterotrifunctional initiator able to initiate the polymerizations of caprolactone (CL) (through two hydroxyl functions, in presence of stannous octoate in a catalytic process) and tert-butylacrylate (tBA) (through a tertiary bromide function, in presence of CuBr/bpy), using a sequential process. After hydrolization, we got the amphiphilic star copolymer (PCL)2-PAA. The amphiphilic copolymer can self-assemble to nanoparticles, used as high loaded drug-carrier.
