Study On The Influence Of The Surface Layer Amino Acids Of Silk Fibroin Fiber To Cell Adhestion And Growth

The cell adhesion of biomaterial refers to the cell adhesion and growth on the surface of biomaterial , so the study on the difference,which is that amino acids composition, structure and hydrophilicity between the surface(or surface layer) and noumenon of biomaterial have influence on the cell adhesion and growth,is very necessary .In this paper,the natural Bombyx mori and tussah fibroin fiber,and the regenerative Bombyx mori and tussah fibroin nanofiber were made as research object,and, the fiber after uncovering surface layer were to contrast.By biochemical detection method,we got the following conclusion:there were not RGD sequence peptides on the surface layer and noumenon of Bombyx mori fibroin fiber, and there were RGD sequence peptides on the surface layer and noumenon of tussah fibroin fiber. By analysis,we can speculated that the ratio of RGD sequence peptides on biomaterial was closely related to the content of aminosuccinic acid and Arginine which compose to RGD sequence peptides and the content ofβ-turn structure where RGD sequence peptides are,etc.In this paper, by cultivating neural stem cells C17.2 and fibroblast L373 in mice ,we fulliy proved that influential factors of cell adhesion and growth on the natural fibroin fiber and the regenerative fibroin nanofiber. By analysis,we can got that , in the view of materialogy, the cell adhesion of biomaterial is related to the content of basic amino acid,β-turn structure and RGD sequence peptides. We can also got that the tussah fibroin fiber contained more basic amino acid than the bombyx mori fibroin fiber, and it has high rate ofβ-turn structure,and ,and existed the RGD sequence peptides.So, at last,we got a conclusion that the cell adhesion of tussah fibroin fiber is better ,and good for cell growth.
