| Objective:1. To investigate the effects of the serum of the patients with APS (of different concentration) on MMP-9 expression of the human umbilical vein endothelial cells (HUVEC). 2. To investigate the effects of fluvastatin (of different concentration) on MMP-9 expression of the human umbilical vein endothelial cells (HUVEC) induced by the serum of the patients with APS.Materials and methods:1. Cell culture and Grouping HUVEC were incubated in vitro and were distributed randomly five groups. They were treated with RPMI1640 culture medium, serum of normal person (25%,50%,100%), serum of the patients with APS (25%,50%,100%), serum of the patients with APS and fluvastatin (2.5,5,10μmol/L), serum of the patients with APS, fluvastatin (2.5,5,10μmol/L) and mevalonate (0.2mmol/L). Different concentration fluvastatin were doped after 1h of mevalonate. Serum of the patients with APS and RPMI1640 culture medium were doped after 1h of fluvastatin. Three observation time points are 12h,24h,48h after admission, on which time point collects the cultured cell.2. The distribution and morphous of HUVEC were observated by inverted phase contrast microscope.3. The concentration of MMP-9 was assayed by enzyme linked immunosorbent assay (ELISA).4. MMP-9mRNA was detected by RT-PCR.5. Incubation of HUVEC with APS serum led to a higher MMP-9 expression compared with incubation with RPMI1640 culture medium or control serum concentration dependently (P<0.05). And the highest point of MMP-9 expression is 24h after admission.2. Incubation of HUVEC with APS serum led to a higher NF-κBp65 expression compared with incubation with RPMI1640 culture medium or control serum (P<0.05).â…¡The effects of fluvastatin on MMP-9 expression of the human umbilical vein endothelial cells (HUVEC) induced by the serum of the patients with APS.1. Comparing with the control group fluvastatin significantly inhibited the expression of MMP-9 and NF-κBp65 concentration dependently (P<0.05).2. Mevalonate inhibited the effects of fluvastatin (P<0.05).Conclusions:1. Incubation of HUVEC with APS serum led to a higher MMP-9 and NF-κBp65 expression, which provides favourable clues to the pathogenesis, clinical reactiveness and prognosis of the DVT&APS.2. Fluvastatin inhibited APS-stimulated MMP-9 expression, which indicats that fluvastatin can protect vascular endothelial cell (VEC) and inhibit thrombosis.3. Fluvastatin inhibited APS-stimulated MMP-9 expression via the NF-κB pathway. |
