Objective: To explore the effects of RNAi targeting IL-1βgene on the production of imflammatory factors IL-1β, TNF-α, IL-6 produced by macrophages. Methods: Three siRNA interferece sequences targetting IL-1βgene were designedand synthesized. RAW264.7 cells were transfected with siRNA mediated by liposome, and the control group was set up. The suppression effect of treatment with siRNA on LPS-stimulated IL-1β, TNF-α, IL-6 secretion in RAW264.7 macrophages was observed by Real-Time PCR and ELISA, and the most effective siRNA sequence was screened.Results: siRNA1 and siRNA2 can suppress the secretion of IL-1βstimulated by LPS in RAW264.7 macrophages on mRNA and protein level. There are no significant deviation in the mRNA expression of TNF-αand IL-6.Conclusions: siRNA targeting IL-1βgene mediated by liposome can silence the expression of IL-1βgene of murine macrophages efficiently.
