Effects Of NF-κB P65 Antisense Oligodeoxynucleotide On The Expression Of Extracellular Matrix Induced By High Glucose In Proximal Tublar Epithelial Cells

Objective To investigate the effects of nuclear factor of kappa B p65 (NF-κB p65) antisense oligodeoxynucleotide (AS-ODN) on high glucose induced expression of NF-κB p65,fibronectin(Fn) and collagenⅢ(ColⅢ) in human proximal tubular epithelial cells (HK-2) under hyperglycemia,so as to explore the role of NF-κB in the renal interstitial fibrillation. Methods The HK-2 cells cultured in vitro were exposed to different concentration of glucose(NG:5.5 mmol/L D-glucose;HG:30mmol/L D-glucose;MG:5.5mmol/L D-glucose+24.5mmol/L mannitol),and different concentration(ODNL:1.2μg/ml NF-κB p65 AS-ODN+30mmol/L D-glucose;ODNH: 1.8μg/ml NF-κB p65 AS-ODN+30mmol/L D-glucose) of AS-ODN/SofastTM was transfected into HK-2 and subcultured under hyperglycemia. NF-κB protein expression in HK-2 cells were measured by immunohischemistry;the concentration of Fn and ColⅢprotein in supernatant of HK-2 cells were detected by ELISA. Results The expression of NF-κB p65 protein in NG and MG groups was mainly in the cytoplasm,while ones in HG group was mainly in the nucleus. The expression of NF-κB p65 protein in HG group increased significantly comparison with that in NG group(P﹤0.01).The concentration of Fn and collagenⅢin HG group significantly higher than in NG group(P﹤0.01). The expression of NF-κB p65 protein in HK-2 cells in ODNL and ODNH groups with transfected NF-κB p65 AS-ODN decreased significantly in comparison with in HG group without ones.The level of Fn and collⅢin supernatant of cultured HK-2 cells in ODNL and ODNH groups were significantly lower than in HG group(P﹤0.05 or P﹤0.01). Conclusion NF-κB AS-ODN can inhibit the expression of FN and ColⅣin proximal tubular epithelial cells induced by high glucose.The inhibitory effect may be mediated by down regulation of NF-κB expression,which may be one of the potential measures to prevent from fibrillation of kidney with diabetic nephropathy.
