Study On Induction Of Apoptosis Of NB4 Cell Line By Citral And Its Mechanis

Objective This study investigates the effect of Citral on acute promyelocytic leukemia cell line NB4 cell growth inhibition and induced apoptosis, try to find the mechanis of induceing apoptosis and lays foundation for further relative research.Methods.1.The NB4 cell shape changes after Citral impact were observed with optical microscopy.2.Using trypan blue staining to observe the growth of NB4 cells and effect of Citral on NB4 cells.3.The effect of different concentrations of Citral and As2O3 on NB4 cell proliferati as well as the role of the same concentration of Citral and As2O3 at different time effects on cell proliferation was determined by CCK-8 assay.4.FCM was used to measure the proportion of early apoptosis under different Citral concentrations and As2O3 or the same concentration of Citral and As2O3 at different time effects on cell.5.The change of mitochondrial membrane potential after Citral impact were detcted by double staining using Rh123/PI.Results Citral significantly inhibits NB4 cell growth.The effect is positively related to the time and dose.After applying 20pμg/ml Citral for 8 hours, optical microscopy observed that the cells show chromatin condensation, marginalization, karyolemma disintegration, chromatin segmentation and apoptotic bodies, and other typical forms of apoptosis.FCM results show that the rate of cell apoptosis increases as Citral concentration and action time increased gradually.Cell mitochondrial membrane potential decreased significantly after NB4 cells effected by Citral.Conclusion Citral can inhibit the NB4 cell growth and proliferation. Its effect is positively relative to the concentration and duration of time.Citral can also induce NB4 cell apoptosis. The percentage of apoptosis is positively related of the concentration and duration of time.The collapse of mitochondrial membrane potential caused by Citral may be one of the mechanisms of apoptosis.
