ObjectTo detect plasma microRNA-21 expression level of patients with hepatocellular carcinoma(HCC)then evaluate its clinical significance as a biochemical index in early diagnosis and prognostis of HCC.MethodSelected 15 HCC patients in the Second Xiangya Hospital, Central South University as the experimental group U, and 10 cases of chronic hepatitis B with or without hepatitis B virus liver cirrhosis as the control group C1, and 10 healthy volunteers as negative control group C2;and also compared the expression level of plasma miR-21 among pre-operation,one week after operation and post-operation 4 weeks paired samples from 10 HCC patients. MiR-21 in the blood plasma was carried out for relatively quantitative analysis by real-time fluorescent quantitative RT-PCR method which was based on 2 (-△△Ct).Results:1.Compared with control group, plasma MiR-21 expressions of group U were significantly higher (p<0.001),which over-expressed 30.58 times more than that of C2 group(p<0.01) and were about 4 times higher than that of in C1 group(p=0.019).2. Plasma MiR-21 expression levels of HCC patients after radical resection decreased progressively(p<0.05).3. Plasma MiR-21 expression levels of HCC patients had no correlation with serum AFP expression level.(r= 0.559,p=0.093>0.05) 4.Plasma MiR-21 expression level in HCC had no correlation with clinical and pathological features including sex, age, tumor size and encapsulation integrity(p>0.05), but it was significantly positive correlated with liver metastasis, liver cirrhosis, tumor thrombus and TNM stage (p<0.05)Conclusion:1.Detection of circulating miR-21 might provide a new complementary biochemical index for diagnosis of early hepatocellular carcinoma.2.MiR-21 expression level in HCC was associated with clinical and pathological features of liver metastasis, liver cirrhosis, tumor thrombus and so on.
