Objective:To detect the expression of vascular endotheial growth factor receptor-3(VEGFR-3,shorter for Flt-4) and D2-40 labeled microlymphtic vessel density(MLVD) in normal cervical, cervical intraepithelial neoplasia(CIN)and invasive cervical sqamous cell carcinoma(CSC). And to assess the relationship of both and clinicopathological parameters and lymphatic metastasis of CSC.And to provide useful marker for clinical forecasting lymphatic metastasis of CSC.Materials and Methods1. Materials89 case of Patients were treated between October 2007 and November 2009 were enrolled in this study.There were 11 case of normal cervical, 15 case of cervical intraepithelial neoplasia(CIN)and 63 case of invasive CSC in them. International Federation of Gynecology and Obstetrics (FIGO) stage were 2 as FIGO stage Ia2,26 as FIGO stage Ib,,8 as FIGO stageⅠb2,25 as FIGO stageⅡa and 2 as FIGO stageⅡb.29 cases were lymphatic metastasis that confirmed by Histological review and 34 Were negative.Based on the analysis of histological grade,6 cases were HG1,32 were HG2, and 25 were HG3.2. Methods Immunohistochemical (IHC) staining with the antibodies against Flt-4 and D2-40 was used to examine the expression of them in normal cervix, CIN and invasive CSC. All statistical calculations were performed using SPSS 17.0.Result1.The expression of Flt-4 in normal cervix, CIN and invasive CSCFlt-4 were mostly localized in the cytoplasm and membrane of the cancer cell in the examined CSC samples and the positively stained cells showed a brown-yellow color.The positive rates were 0%(0/11),6.67%(1/15) and74.6% (47/63) in normal cervix, CIN and invasive CSC samples, respectively. There were significant differences of positive expressive rates of Flt-4 among normal cervix, CIN and invasive CSC samples. (x2=37.20, P=0.000).There was no significant difference in the two-two comparisons between the normal cervix and CIN (exact P =0.577).The rest were significant differences each other.(all P= 0.000) D2-402.2. D2-40 labeled MLVD in normal cervix, CIN and invasive CSC D2-40 were mostly localized in the cytoplasm of the positive lymphatic endothelial cells and were stained in a brown-yellow color. It labeled MLVD were found to be (1.55±1.04), (7.40±2.87) and (17.14±5.53) in normal cervix, CIN and invasive CSC,respectively.
