Study On The Secretion And Influence Factors Of Immunoreactive Growth-hormone In Systemic Lupus Erythematosus Patients

Objective:To investigate the role of mmunoreactive growth hormone(irGH) in the pathogenesis of SLE,Study the secretion of irGH in peripheral blood mononuclear cells (PBMC)of SLE patients, and factors which influence its secretion.Method:39 SLE patients was collected(19 active stage,20stable stage)from both outpatient and inpatient of dematology department in Huashan Hospital,and 20 normal people matched with gender and age as control. Double-antibody sandwich ELISA was used to determine the cellular supermatant contents of irGH and the influence of different intervention factors:IL-4, IFN-y,prolactin, Methyllprednisolone.Result:The secretion of irGH was significantly more in SLE patients both in active stage (3148.565±359.2037 ng/L) and stable stage(2855.348±378.4728ng/L) than normal control(2568.324±255.4337) (p<0.OO1,P=0.0128),and active stage patients were also more than stable stage patients (p=0.0178).The level of irGH secretion was related with SLEDAI,serum dsDNA and anti-nucleosome antibodies in active stage patients (p<0.001, p=0.001, p=0.0263).Furthermore,the level of irGH secretion is also related to IgG and dsDNA secreted by PBMC of SLE patients cultured in vitro (P<0.01).IL-4 and prolatin up-regulated the secretion of irGH,and methylpredinidone down-regulate the secretion of irGH,but the influence of IFN-γon the secretion of irGH hadn't been observed.Conclusion:The secretion of irGH of SLE patients both in active stage and stable stage irGH was abnormally raised,moreover, patients in active stage higher than in stable stage. The level of irGH secretion was related with SLEDAI,serum dsDNA and anti-nucleosome antibodies in active stage patients.Furthermore,the level of irGH secretion is also related to IgG and dsDNA secreted by PBMC of SLE patients cultured in vitro (P<0.01).IL-4 and prolatin up-regulated the secretion of irGH,and methylpredinidone down-regulate the secretion of irGH,but the influence of IFN-γon the secretion of irGH hadn't been observed.Maybe irGH play an important role in the pathogenesis of systemic lupus erythematosus.
