Objective To investigate the role and mechanism of interleukin-1βon fibroblast synthesis of COL1A2 and matrix metalloproteinase-3, and explore the impact of interleukin-1βon the formation of epidural scar.Methods The morphology of NIH3T3 cells were observed by inverted microscope; The NIH3T3 cells were divided into three groups:IL-1βgroup, IL-1β+SB202190 group and control group. After serum-free culture for 20 hours, The IL-1B group was treated by IL-1B (10ng/ml) and IL-1β+SB202190 group was treated by the SB202190 (10μmol/L) for 1 hour before treated with IL-1β(10ng/ml) and the control group was treated with 2% serum. Then those 3 groups were culturing for 24 hours under standard conditions (37℃in a humidified atmosphere flushed with 5% CO2 in air). The expression of MMP-3 and COL1A2 gene was examined by RT-PCR and Western blotting.Results (1)The expression of COL1A2 mRNA among these three groups had significant difference by RT-PCR (F=55.67,P<0.01). Between the IL-1B group and the IL-1β+ SB202190 group or the control group, there was a significant difference in the expression of COL1A2 mRNA(P<0.01). The IL-1β+SB202190 group's expression of COL1A2 mRNA was lower than the control group, but there's no significant difference(P>0.05).Detections of COL1A2 protein were performed by Western blotting. There were significant difference among these three groups (F=251.01,P<0.01). The expression of COL1A2 protein in the IL-1B group had significant difference compared with the other two groups(P<0.01). But the comparation between the IL-1β+SB202190 group and the control group had no significant difference (P>0.05).(2)The expression of MMP-3 was significantly increased in the IL-1βgroup than in the control group (P<0.01). The expression of MMP-3 in the IL-1β+SB202190 group was significantly reduced compared with IL-1B group (P<0.01), but no significant difference was noted between the IL-1β+SB202190 group and the control group (P>0.05).Conclusions IL-1βcould inhibit the expression of COL1A2, and stimulate the expression of MMP-3 in NIH3T3 cells which might inhibit the formation of epidural scar. Besides, we found that the p38 pathway played an major role in this process.
