| Objective:The study was designed to observe the combination effect of celecoxib and 5-fluorouracil (5-FU) in human gastric carcinoma cell line SGC7901 in vitro. Methods: The combination effect and the interaction between celecoxib and 5-FU on SGC7901 cells were evaluated according to the median-effect principle and MTT. Results:When celecoxib or 5-FU was used alone, the results showed that both agents had the concentration-dependent inhibitory effect. The celecoxib concentration inhibiting 50% (IC50) of the growth of SGC7901 cells was 37.527μmol. L-1, and the 5-FU concentration inhibiting 50% of the growth of SGC7901 cells was 133.487μmol.L-1. When using celecoxib and 5-FU at the same time, we discover that the IC50 of celecoxib and 5-FU is 10.448μmol.L-1 and 50.151μmol.L-1. It ws accelerated inhibit effect when the two agents given concurrently. By median-effect principle analysis, the combined effect of the two agents given concurrently was synergistic. The synergistic effect was shown no relation with the different given orders. Conclusion:The combination effect of celecoxib and 5-FU given concurrently is synergistic. The results have clinical value in the treatment of gastric carcinoma. |
