Objective In this study we applied siRNA directed against EGFR gene to inhibit expression of EGFR gene in ovarian epithelial carcinoma cisplatin-resistance cell line SKOV3/DDP, discussed effects of RNA interference against EGFR gene on increasing the apoptosis of SKOV3/DDP. Methods pEGFR-shRNA was synthesized in vitro by the genetic recombination and transfected into SKOV3/DDP by liposome. RT-PCR and ICC were performed respectively to detect the inhibitive effect of RNA interference on mRNA level and protein level. The apoptotic rates and cell cycle of SKOV3/DDP cells were detected by flow cytometry after having been treated with cisplatin. Results DNA sequencing confirmed that the insertion sequence was completely correct. The expression of EGFR mRNA of sequence specific pEGFR-shRNA was obviously down-regulated and the protein expression was also obviously reduced. Flow cytometric analysis showed that cell cycle distribution was changed and the ratio of apoptosis significantly increased after transfection. Conclusion The EGFR specific shRNA expression vector can effectively suppress the expression level of EGFR gene and protein of SKOV3/DDP cells and induce cells apoptosis.
