With the widely use of antibiotics, staphylococcus aureus's drug resistance has been an increasing problem, such as drug-resistant strains and multiple resistant strains. In order to solving this problem, to develop drug-resistant inhibitor has become the focus of attention. The traditional Chinese medicines have relatively minor toxic side effects and more effective ingredients. Therefore, the research integrated various aspects of information, trying to select the drug resistance inhibitors from traditional Chinese medicines and preliminary study the resistance mechanism of inhibition.Through microscopy, biochemical characteristics and animal inoculation methods, the result of this study showed that the bacterial strains belonged to gram-positive staphylococcus aureus. not mixed bacterium pollute,The bacteriostatic strengths that were related to 14 kinds of antimicrobial attending to 5 strains of bacteria were tested by drug susceptibility pieces and trace dilution determination methods. Refer to NCCLS judgment standard, the results confirmed that staphylococcus aureus resistance to 9 species antibiotics. through artificially inducing norfloxacin resistance to staphylococcus aureus, the result resistant drug rate could be improved.11 kinds of effective constituents of traditional Chinese medicine were extracted by the water extraction and alcohol extraction methods, using drug susceptibility pieces and trace dilution determination methods to detect the in vitro bacteriostasis about drug-resistant bacteria. Furthermore, the restrain experiments were implemented about the bacteriostatic strong resistance of traditional Chinese medicine. The results indicated that coptis's extract appeared the strongest antimicrobial resistance and inhibition. The diameter of bacteriostatic circle was 27.25±2.1 minimum, inhibitory concentration 3.9 mg/ml, and highest inhibition rate 27%. Testing in oxford cup test method, the result showed that the extract of coptis could enhance the antibiotic sensitivity for bacteria. Using the fluorescence detector, with the 468nm emission wavelength and 282nm stimulate wavelength we tested the accumulated concentration of norfloxacin. The result indicated that the accumulated concentration of medicine norfloxacin within the experiment strains which treated with coptis extract was clear high than blank space (P<0.01). The result shows that it to have promote in vivo bacterium medicine accumulated effect.Using SDS-PAGE electrophoresis and spectrophotometry, we detected tropina variety and the enzymatic activity in extracellular fluid before and after coptis extract disposition. The result showed that some proteins which molecular weights were in 35KDa-38KDa rebound expressed after coptis extract treated, but the extracellular fluid enzymic activities of resistant staphylococcus aureus had no significant difference (P>0.05). it can inter there no influence on resistant the medicine golden yellow S.aureus's cell membrane's permeability.Above results indicated:the above of drug-resistant bacteria coptis inhibition of various chemical composition, the traditional Chinese drug-resistant inhibitors of developing is to alleviate the increasingly serious bacterial resistance of effective channel.
