Objective:To investigate the expression and significance ofβ-catenin, cyclooxygenase-2(COX-2) and prostaglandinE2(PGE2) in the dimethylhy-drazine (DMH) induced colon carcinogenesis of mucosa in rats.Methods:90 samples of Wistar rats were divided into 2 groups at random, including 10 controls and 80 DMH-induced colon carcinogenesis models. COX-2 andβ-catenin mRNA's and protein's expression in the colorectal mucosal tissues of rats were detected by real-time quantitative reverse transcription polymerase chain reaction (Real-time PCR) and Western-blot, respectively. The expression of PGE2 in the colorectal cancer was detected by Elisa.Results:35 rats were induced to develop adenocarcinomas, and 8 samples of adenoma with atypical hyperplasias, there was no tumor in control group. The mRNA and protein ofβ-catenin in the colorectal cancer and the adenoma with atypical hyperplasia tissue were all higher than normal mucosa (P<0.01), and the cancers were higher than adenoma with atypical hyperplasia tissue (P<0.01); The expressions of COX-2 mRNA and protein in the colorectal cancer tissues were higher than that in the adenoma with atypical hyperplasia and the normal mucosa tissue (P<0.05), and the adenoma with atypical hyperplasia tissue were higher than normal mucosa tissue (P<0.05). The expressions of PGE2 in colorectal cancer were higher than adenoma with atypical hyperplasia tissue and the normal mucosa tissue, the adenoma with atypical hyperplasia tissue were higher than the normal mucosa tissue (P<0.05).Conclusions:The expresssions of B-catenin, COX-2 and PGE2 were higher and higher in the formative processes of colorectal cancer.
